      目的 通过对于骨髓移植后甘油注射所致肾小管损伤小鼠使用川芎嗪,粒巨噬细胞集落刺激因子(GM-CSF ),观察促进骨髓基质细胞增殖的因素在骨髓干细胞对于肾小管损伤后参与修复的影响。方法 绿色荧光蛋白标记(GFP)的C57BL/6转基因小鼠提供骨髓,同种无荧光标记的C57BL/6小鼠分为生理盐水组(n=20)、20%川芎嗪干预组(n=20)、GM-CSF组干预组(n=20)进行血常规、肾功能、流式细胞仪以及肾脏病理检测,并采用荧光组织化学、免疫组织化学等方法观察绿色荧光在骨髓移植小鼠肾脏的分布及数量。结果 经过骨髓移植以及甘油注射肾小管损伤后,三组在流式细胞仪结果中显示干预组GFP阳性细胞偏高(P<0.05),而病理结果显示:川芎嗪组病理评分结果较其他两组高(P<0.05),而GM-CSF有双向的表现。免疫组化显示干预组均有外源性的肾小管上皮细胞的增多(P<0.05)。结论 川芎嗪和GM-CSF均可一定程度增强骨髓干细胞对肾小管损伤的修复作用。

      Objective To advanced demonstrate bone-marrow-derived mesenchymal stem cells can differentiate into renal tubular epithelial cells and discuss the factor of accelerating hematopoietic reconstitution effect on the repair of tubular injury by bone marrow transplantion mice perform glycerol injection.Methods Transgenic C57BL/6 mice labeled with GFP as bone maeeow donors,C57BL/6 mice,recipients of bone marrow transplantion,were divided into three groups:(1)normal saline group;(2)20%Ligustrazine group,(3)GM-CSF group.All groups procee blood rountine,renal function,flow cytometry of bone marrow cells,and each side kidney histology,distribution and amount of green fluorescence in kidney of recipient mice observed by fluorescence histochemistry,immunohistochemistry means.Results Flow cytometry of bone marrow cells show the 2,3 groups more increase than the 1 group,the 2 group can enhance the recovery of renal injury,the 3 group show a “two-way”influence in the recovery of renal injury.In the fluorescence histochemistry and immunohistochemistry,the 2,3 groups show have a few increase in the number of ectogenesis(GFP+) renal tubular epithelial cells.Conclusion Ligustrazine or GM-CSF can enhanced the repair of tubuLar injury in glycerol injection injury mice by bone-marrow-derived mesenchymal stem cell.
