      目的 观察橙皮苷对化学性肝纤维化大鼠α-平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)表达的影响,探讨其可能的作用机制。方法 采用50%四氯化碳(carbon tetrachloride,CCl4)每周2次,连续12周皮下注射的方法,诱导肝纤维化大鼠模型。造模后第7周,每天按50,100,200 mg·kg-1灌胃给予不同剂量的橙皮苷(hesperidin,HDN)和秋水仙碱(0.1 mg·kg-1),共6周。末次给药后,禁食8 h,处置大鼠,分光光度法检测血清丙氨酸转氨酶(alanine aminotransferase,ALT)、天冬氨酸转氨酶(aspartate aminotransferase,AST)和肝组织羟脯氨酸(hydroxyproline,Hyp)的含量;取固定部位肝脏组织,HE和Masson染色观察肝组织病理损伤程度,免疫组化和RT-PCR技术检测肝组织中α-SMA蛋白与mRNA的表达。结果 与模型组相比,HDN (100,200 mg·kg-1)组不仅可改善大鼠肝纤维化损伤程度,降低ALT、AST、Hyp含量(P<0.05或P<0.01),还可减少肝组织中α-SMA蛋白与mRNA的表达(P<0.05或P<0.01)。结论 HDN对化学性肝纤维化大鼠具有很好的保护作用,其机制可能与抑制肝星状细胞的活化、增殖有关。

      Objective To observe the effect of hesperidin (HDN) on α-smooth muscle actin (α-SMA) expressions in livers of hepatic fibrosis rats and to explore the underlying mechanisms. Methods SD rats were randomized into control group, model group, HDN (50,0, 200 mg·kg-1) groups and colchicines (0.1 mg·kg-1) group. Rat with liver fibrosis was induced by hypodermic injection carbon tetrachloride (CCl4), twice a week for 12 weeks. Then different doses of HDN and colchicine were administrated via gavage once a day from week 7 to 12 after liver fibrosis was induced. At the end of experiment, the levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), and hydroxyproline (Hyp) were measured by chromatometry. Meanwhile, liver samples were taken to examine the degree of liver fibrosis by HE and Masson. Moreover, the expressions of α-SMA in liver were observed by immunohistochemistry and transcriptase-polymerase chain reaction (RT-PCR). Results As compared with the model group, HDN (100,0 mg·kg-1) not only obviously improved histopathological change, but also degraded the contents of ALT, AST and Hyp(P<0.05 or P<0.01). Meanwhile, HDN could effectively prevent the protein and mRNA expression of α-SMA(P<0.05 or P<0.01). Conclusion HDN has a powerful protective effect on liver fibrosis in rats, which might be partly associated with its effects on inhibiting the activation and proliferation of HSC.
