      目的 研究熊果酸(UA)对人上皮性卵巢癌SKOV3细胞增殖和凋亡的影响,并探讨其可能的作用机制。方法 将处于对数生长期人上皮性卵巢癌SKOV3细胞随机分为5组:正常对照组、熊果酸(40、20、10 mg·L-1)干预组和顺铂(40 mg·L-1)干预组(阳性对照组)。药物干预48 h后,通过四甲基偶氮唑蓝(MTT)比色法测定细胞增值抑制率;通过流式细胞仪分析细胞周期的改变、观察细胞凋亡并计算细胞凋亡率;通过RT-PCR技术检测细胞Bax mRNA和bcl-2 mRNA表达并计算Bax/bcl-2表达比值,Western blot法检测细胞半胱氨酸蛋白酶-3(caspase-3)蛋白表达并进行半定量分析。结果 较正常对照组,熊果酸(40、20 mg·L-1)干预组细胞增殖抑制率显著升高(P<0.01),细胞周期被阻滞于G2/M期,细胞凋亡率显著升高(P<0.01);熊果酸(40、20 mg·L-1)干预组细胞中bcl-2 mRNA表达显著下调、Bax mRNA表达显著上调,Bax/bcl-2表达比值显著升高(P<0.01),caspase-3蛋白表达显著上调(P<0.01)。结论 熊果酸具有抑制人上皮性卵巢癌SKOV3细胞增殖并促进其凋亡的作用,其作用机制可能与熊果酸能够有效下调bcl-2 mRNA表达、上调Bax mRNA表达、提高Bax/bcl-2表达比值以及上调caspase-3蛋白表达有关。

      Objective To investigate the effects and mechanism of ursolic acid(UA) on the proliferation and apoptosis of epithelial ovarian SKOV3 cancer cell. Methods The Logarithmic epithelial ovarian cancer SKOV3 cells were randomized into five groups:normal control group, UA(40、20、10 mg·L-1) groups and Cisplatin 40 mg·L-1 group. 48 h later, the cellular growth inhibition rate was calculated by MTT, cell cycle and apoptosis rates were analyzed by flow cytometry, the expression of Bax mRNA and bcl-2 mRNA were detected by RT-PCR, the level of caspase-3 protein expression was detected by Western blot. Results Compared with normal control group, the cellular growth inhibition rates of UA(40、20 mg·L-1) groups were significantly increased(P<0.01); the cell cycle was arrested at G2/M phase, the apoptosis rate was significantly increased(P<0.01); the expression of bcl-2 mRNA was significantly decreased, while the expression of Bax mRNA was significantly increased, and the ratio of Bax/bcl-2 was significantly increased(P<0.01); the expression of caspase-3 protein was significantly increased (P<0.01). Conclusions UA could effectively inhibit epithelial ovarian SKOV3 cancer cell proliferation and promote its apoptosis, which was perhaps related to its effects of downregulating the expression of bcl-2 mRNA, raising the expressions of Bax mRNA and caspase-3, and enhancing the ratio of bcl-2/Bax.
