      目的 应用FlowCytomix检测类风湿关节炎患者(RA)细胞因子表达谱,探讨其在RA发病及治疗中的作用。方法 分离RA患者及健康对照者外周血单个核细胞(PBMCs),经PHA刺激后,收集细胞培养上清,FlowCytomix检测上清中细胞因子浓度。结果 与健康对照组相比,RA患者IL-17、IL-6、IL-1β、TNF-α水平均显著升高(P<0.05),但IFN-γ水平显著降低(P<0.05),IL-10水平无明显改变。RA患者疾病活动组IL-17、IL-6、IL-1β、TNF-α水平显著高于缓解组(P<0.05),但IFN-γ水平低于缓解组(P<0.05);IL-17、IL-6、IL-1β、TNF-α、IFN-γ水平与DAS28评分显著相关(P<0.05)。结论 FlowCytomix多细胞因子检测系统可特异、灵敏地检测RA患者PBMCs中细胞因子产生,RA患者PBMCs中IL-17、IL-6、IL-1β、TNF-α水平反映了其关节的炎症程度及活动状态。

      Objective To detect cytokine profile of patients with rheumatoid arthritis (RA) by FlowCytomix. Methods Peripheral blood mononuclear cells (PBMC) were isolated from healthy controls and RA patients by Ficoll-Hypaquedensity gradient centrifugation, then the cells were stimulated with phytohaemagglutinin (PHA) and the supernatants were collected to quantitatively measure cytokine production by FlowCytomix. Results Compared with control group, the concentrations of IL-17, IL-6, IL-1β and TNF-α were significantly increased but the level of IFN-γ was decreased. The concentration of IL-10 was not significantly changed. The levels of IL-17, IL-6, IL-1β, and TNF-α were higher in active RA than those of remission, whereas IFN-γ level was high in remission. IL-17, IL-6, IL-1β, TNF-α, and IFN-γ levels and DAS28 score showed significant correlation.Conclusions Multiplex cytokine profiling systems of FlowCytomix can be applied to specific and sensitive quantification of cytokines of PBMCs from RA patients, and the levels of IL-17, IL-6, IL-1β, and TNF-α in PBMCs reflect the degree of inflammation in joints and the disease activity.
