      目的 观察脂多糖(LPS)对类风湿关节炎(RA)患者外周血单核细胞Toll样受体4(TLR4)、髓样分化因子88(MyD88)蛋白表达的影响。方法 采用活动期RA患者外周血进行单核细胞的分离培养,并与健康志愿者做对照。分为以下5组观察:健康对照组、健康+LPS组、RA对照组、RA+ LPS组、RA+LPS+TAK-242组,并采用Western blot法检测各组TLR4、 MyD88蛋白的表达。结果 健康+LPS组或RA+LPS组TLR4、MyD88蛋白表达与健康对照组或RA对照组比较均明显升高(均P<0.01);RA对照组或RA+LPS组TLR4、MyD88蛋白表达与健康对照组或健康+LPS组比较均明显升高(均P<0.01);RA+LPS+TAK-242组TLR4、MyD88蛋白表达与RA+LPS组比较明显降低(均P<0.01)。结论 RA活动期单核细胞可能存在TLR/MyD88信号通路的激活,且对LPS的刺激反应性增强。

      Objective To observe the effect of lipopolysaccharide(LPS) on expression of TLR4 and MyD88 protein in peripheral blood monocytes cells of rheumatoid arthritis(RA).Methods Peripheral blood monocytes cells of active RA patients were isolated and cultured,healthy volunteers as control.Specimen were assignedinto the following five groups:the healthy control group,health + LPS group,RA control group,RA + LPS group,and RA + LPS + TAK-242 group.The expressions of TLR4 and MyD88 protein were detected by Western blot.Results The expressions of TLR4,MyD88 protein in health + LPS group or in RA + LPS groupwere significantly higher than those in healthy control group or in RA control group(all P<0.01).The expressions in RA control group or in RA + LPS group were significantly higher than those in healthy control group or health + LPS group(all P<0.01).The expressions in RA + LPS + TAK-242 group were significantly lower than those in RA + LPS group(all P<0.01).Conclusion Monocytes cells of active RA patients may haveTLR / MyD88 signaling pathway activation and their reactivity to LPS stimulation is increased.
