      目的 研究丹红注射液对小鼠感染人巨细胞病毒(HCMV)病毒性肝炎的干预机制,为临床治疗HCMV提供理论依据。方法 BALB/c 品系小鼠80只,随机数字表编号法分对照组、模型组、丹红注射液A、B、C组(腹腔注射丹红注射液,剂量分别为0.5、1、2 mg·10 g-1·d-1),对照组、模型组用生理盐水干预治疗,连续28 d。28 d后尾静脉采血,放免法检测小鼠血清肝纤维化血清学指标[Ⅳ型胶原(cⅣ)、层黏连蛋白(LN)、转化生长因子-βl(TGF-βl)、透明质酸(HA),Ⅲ型前胶原(PC-Ⅲ)],用小鼠尾静脉血血清检测肝功能五项[天冬氨酸氨基转移酶(AST)、胆汁酸(TBA)、总胆红素(TBil)、谷氨酰转移酶(GGT)、丙氨酸氨基转移酶(ALT)];检测肝脏组织中肝微粒体细胞色素P450 亚型酶[CYP2E1、CYPIA2、CYP3A4、CYP2D6、CYP2C9]活性。结果 与模型组比较,丹红注射液B、C组治疗28 d后,P450 亚型酶CYP2E1、CYPIA2、CYP3A4酶活性、血清肝纤维化血清学指标cIV、TGF-βl、PC-Ⅲ及肝功能AST、ALT均有不同程度降低(P<0.05),均差异有统计学意义。丹红注射液A组各指标间比较,差异无统计学意义,B、C组组间比较,差异无统计学意义(P>0.05)。结论 丹红注射液可抑制小鼠P450 亚型酶活性,降低药物代谢时对肝脏的毒性,并通过降低TGF-βl而促进肝功能恢复,对小鼠感染,HCMV病性肝炎有明确的治疗作用。
英文摘要:
      Objective To explore the intervention mechanism of Danhong Injection on mice infected with human cytomegalovirus(human cytomegalovirus,HCMV)viral hepatitis,to provide theoretical basis for clinical treatment of HCMV.Methods Eighty BALB/c strain mice were randomized into control group,model group,Danhong Injection A,B,C group,by intraperitoneal injection of Danhong Injection(0.5,1,2 mg·10 g-1·d-1).The control group and model group received normal saline intervention therapy for 28 days.28 days later,blood samples were collected from tail vein for immunoassay detection of serum markers of liver fibrosis in mice [Ⅳ collagen(cⅣ),laminin(LN),transforming growth factor beta 1(TGF-β1),hyaluronic acid(HA),type Ⅲ procollagen(PC-Ⅲ)],detection of liver functions in mice [aspartate aminotransferase(AST),bile acid(TBA),total bilirubin(TBil),gamma glutamyl transferase(GGT)alanine aminotransferase(ALT)],and the activities of liver microsomal cytochrome P450 isoforms [CYP2E1,CYPIA2,CYP3A4,CYP2D6 AND CYP2C9].Results Compared with the model group,28 days after the treatment,activities of CYP2E1,CYPIA2 and CYP3A4,cIV,TGF-βl,PC-III,AST and ALT were all decreased in Danhong Injection Band Cgroup with significant differences(P<0.05).There was no statistically significant difference among Danhong Injection A,B and Cgroup(P>0.05).Conclusions Danhong Injection can inhibit the activity of P450 isoforms in mice,decrease toxicity to liver during drug metabolism and promote the recovery of liver function by reducing TGF-βl,which achieves obvious efficacy on mice-infected human cytomegalovirus hepatitis.
