| 目的 探讨血清miRNA-125b与miRNA-21联合检测前列腺癌患者的临床诊断价值。 方法 选择崇州市人民医院2014年1月至2016年1月收治的65例前列腺癌患者为研究对象,另选择同期良性前列腺增生患者65例、健康体检者65例作为对照,通过实时荧光定量PCR(RT-qPCR)检测血清miRNA-125b与miRNA-21表达量,并进行统计学分析。 结果 前列腺癌患者血清miRNA-125b、miRNA-21水平显著高于良性前列腺增生和健康体检者(P＜0.05),而良性前列腺增生和健康体检者之间比较差异无统计学意义(P＞0.05)；miRNA-125b在年龄＞60岁、Gleason评分＞7、PSA≥10 μg·L-1、T3~4期、远处转移患者高表达(P＜0.05),miRNA-125b在Gleason评分＞7、PSA≥10 μg·L-1、T3~4期患者高表达(P＜0.05)；miRNA-125b诊断前列腺癌的ROC曲线下面积(AUC)为0.799(95%CI:0.650~0.949),miRNA-21的AUC为0.733(95%CI:0.588~0.879),而二者联合诊断的AUC为0.904(95%CI:0.798~0.982)(P＜0.05)。 结论 血清miRNA-125b与miRNA-21在前列腺癌患者血清高表达,且与患者病理特征有一定关系,二者联合检测可提高对前列腺癌的诊断效率。 |

| Objective To investigate the diagnostic value of combined detection of serum miRNA-125b and miRNA-21 in prostate cancer. Methods 65 prostate cancer patients enrolled in Chongzhou Hospital from January 2014 to January 2016 were selected as the research object,65 patients with benign prostatic hyperplasia and 65 healthy persons as the control.The expression of miRNA-125b and miRNA-21 in serum were detected by RT-qPCR and statistical analyzed. Results The level of serum miRNA-125b and miRNA-21 in patients with prostate cancer was significantly higher than that in benign prostatic hyperplasia and healthy persons(P＜0.05),however,there is no difference between benign prostatic hyperplasia and healthy persons(P＞0.05).MiRNA-125b is highly expressed in patients with aged over 60,Gleason score ＞7,PSA ≥ 10 μg·L-1,T3-4 stage and distant metastasis(P＜0.05),miRNA-21 is highly expressed in patients with Gleason score ＞7,PSA ≥ 10 μg·L-1,T3-4 stage(P＜0.05).The area under of the ROC curve for miRNA-125b diagnosis of prostate cancer was 0.799 (95 % CI:0.650-0.949) and the miRNA-21 was 0.733 (95 % CI:0.588-0.879),the area under foe combined detection of the two miRNAs was 0.904 (95 % CI:0.798-0.982). Conclusion High expression of serum miRNA-125b and miRNA-21 in the serum of prostate cancer patients are correlated with characteristics of patients,combined of which can improve the efficiency of diagnosis of prostate cancer. |
