      目的观察干扰ROCK1基因对鼻咽癌的细胞侵袭影响。方法 收集2016年1—12月临沂市河东区人民医院30例鼻咽癌病人临床鼻咽癌标本及同期鼻咽癌的癌旁组织,通过免疫组织化学法(免疫组化)检测ROCK1的表达;培养鼻咽癌细胞株6-10B和永生化的鼻咽上皮细胞NP69,蛋白质印迹法(Western Blot)方法检测ROCK1的表达;使用ROCK1的抑制剂Y-27632处理6-10B细胞株,通过Transwell小室实验,检测鼻咽癌的细胞侵袭。结果 免疫组化结果显示,30例鼻咽癌组织中,ROCK1有18例阳性,12例阴性;而对照组中只有2例阳性,鼻咽癌组的ROCK1明显高表达,与对照组相比差异有统计学意义(χ2 =19.20,P<0.000 1);Western Blot检测结果显示,6-10B组的ROCK1表达明显高于NP69细胞组;侵袭实验结果显示,未处理组为(171.67±17.89),明显高于10 μM组(120±19.07)和30 μM组(67.33±13.58),差异有统计学意义(P<0.05)。结论 ROCK1基因与鼻咽癌的细胞侵袭有关。

      To observe the effects of the interfering ROCK1 gene on the cell invasion of nasopharyngeal carcinoma.Methods Clinical samples of 30 patients in Linyi Hedong District Peoples Hospital from January to December 2016 were collected,which nasopharyngeal carcinoma tissue and non nasopharyngeal carcinoma tissue were assigned into the experimental group and the control group.The ROCK1 expression of clinical samples was detected by immunohistochemistry.Nasopharyngeal carcinoma cells line 6 10B and immortalized nasopharyngeal epithelial cells NP69 were cultured.The ROCK1 expression level in cells line was detected by Western blot.After treated with Y 27632 (10 μM and 30 μM),cell invasion was detected by transwell assay.Results Immunohistochemical results showed that 18 of 30 cases of nasopharyngeal carcinoma were positive and 12 cases were negative,while only 2 cases were positive in the control group.Compared with the control gro up,the ROCK1 expression level of nasopharyngeal carcinoma group was significantly higher by immunohistochemical assay (χ2=19.20,P<0.000 1).Western blot assay results showed that ROCK1 expression of 6 10B group was higher than that of NP65 cell group.Transwell assay showed that the cell number of untreated group (171.67±1789) was higher than (120±19.07) of Y 27632 (10 μM) group and (67.3313.58) of Y 27632 (30 μM) group with statistically significant difference (P<0.05).Conclusion ROCK1 gene is associated with the cell invasion of nasopharyngeal carcinoma.
