      目的探究 TPX2和 Aurora?A蛋白在宫颈鳞癌组织中的表达水平及预后意义。方法回顾性选取 2011年 6月至 2013年 6月徐州市中心医院 85例宫颈鳞癌、 30例宫颈上皮内瘤变( CIN)Ⅱ~Ⅲ、30例 CIN Ⅰ和 20例慢性宫颈炎四组病人为研究对象。运用免疫组织化学方法检测 TPX2和 Aurora?A蛋白在各组宫颈组织中的表达情况;本课题组对宫颈鳞癌病人进行随访,采用 Kaplan?Meier法以及 Cox比例风险回归模型分析 TPX2和 Aurora?A蛋白表达水平与宫颈鳞癌病人总生存期的关系。结果 TPX2蛋白在宫颈鳞癌、 CIN Ⅱ~Ⅲ、CIN Ⅰ和慢性宫颈炎四组病人宫颈组织中的表达阳性率分别为 60.0%、50.0%、6.7%、5.0%, Aurora?A蛋白在四组病人宫颈组织中的表达阳性率分别为 63.5%、53.3%、10.0%、5.0%。TPX2和 Aurora?A的蛋白表达水平在宫颈鳞癌和 CIN Ⅱ~Ⅲ宫颈组织中显著高于 CIN Ⅰ和慢性宫颈炎( P<0.05)在宫颈鳞癌组与 CIN Ⅱ~Ⅲ组、 CIN Ⅰ组与慢性宫颈炎组之间差异无统计学意义( P>0.05)。相关分析显示 TPX2与 Aurora?A的,蛋白表达水平呈正相关( r=0.229,P=0.035)。 Kaplan?Meier生存分析显示, TPX2和 Aurora?A蛋白阳性表达病人的总生存期显著低于阴性表达病人( P<0.05)。多因素 Cox回归分析显示, TPX2和 Aurora?A蛋白可作为宫颈鳞癌病人预后的独立危险因素[风险比( HR)=2.449,95%置信区间( CI):1.056~5.676,P=0.037;HR=2.354,95%CI:1.182~4.688,P=0.015]。结论宫颈鳞癌组织中 TPX2和 Aurora?A蛋白表达水平增高,其阳性表达与宫颈鳞癌的预后相关,有望成为指导临床治疗和预后的分子靶标。

      Objective To investigate the expression level and prognostic significance of TPX2 and Aurora?A protein in cervicalsquamous cell carcinoma.Methods Eighty?five patients with cervical squamous cell carcinoma,30 patients with cervical intraepi? thelial neoplasias(CIN)Ⅱ?Ⅲ,30 patients with CIN Ⅰ,and 20 patients with chronic cervicitis in Xuzhou Central Hospital fromJune 2011 to June 2013 were selected retrospectively.Immunohistochemistry was used to detect the expression of TPX2 and Aurora?A protein in cervical tissues of each group.Patients were followed up by ourselves,Kaplan?Meier method and Cox proportional haz?ards regression model were used to analyze the relationship between TPX2 and Aurora?A expression and the overall survival of cer?vical squamous cell carcinoma patients.Results The positive rates of TPX2 expression in cervical tissues of the four groups(cervi? cal squamous cell carcinoma,CIN Ⅱ?Ⅲ,CIN Ⅰ,chronic cervicitis)were 60.0%,50.0%,6.7% and 5.0%,respectively.The positive rates of Aurora?A expression in cervical tissues of the four groups were 63.5%,53.3%,10.0% and 5.0%,respectively.The proteinexpression levels of TPX2 and Aurora?A in cervical squamous cell carcinoma and CIN Ⅱ?Ⅲ patients were significantly higher thanthat in CIN Ⅰ and chronic cervicitis(P<0.05).However,there were no significant difference between the cervical squamous cell carcinoma group and the CIN Ⅱ?Ⅲ group,the CIN Ⅰ group and the chronic cervicitis group(P>0.05).The protein expression levels of TPX2 and Aurora?A were positively correlated(r=0.229,P=0.035).Kaplan?Meier survival analysis showed that patientswith positive expression of TPX2 and Aurora?A had shorter overall survival than patients with negative expression(P<0.05).Multi? variate Cox regression analysis revealed that TPX2 and Aurora?A could be used as independent risk factors for prognosis in pa?tients with cervical squamous cell carcinoma[hazard ratio(HR)=2.449,95% CI:1.056?5.676,P=0.037;HR=2.354,95% CI:
