      目的建立高效液相色谱法测定人血清中拉莫三嗪浓度方法,并将该方法运用到临床工作中。方法血清样本采用二氯甲烷进行萃取。以艾司唑仑为内标物。采用 Inertsil ODS?3色谱柱( 4.6 mm×250 mm,5 μm);流动相:甲醇∶水= 75∶25;流速: 1.0 mL/min;检测波长: 225 nm;柱温: 25 ℃;进样量: 10 μL。结果拉莫三嗪线性回归方程: Y=9.934 4X+0.808 3,r= 0.999 8(n=7);拉莫三嗪血药浓度在 1.00~18.00 μg/mL范围内线性关系良好;日内精密度、日间精密度和稳定性均符合相关要求(相对标准偏差小于 15%);拉莫三嗪 3.0 μg/mL、9.0 μg/mL以及 18 μg/mL浓度的加样回收率均大于 96%,提取回收率均大于 72%。结论该方法操作简便、快速、准确,可用于拉莫三嗪治疗药物监测。推荐拉莫三嗪治疗血药浓度为 3~14 μg/mL,可为临床个体化、合理用药提供参考。
英文摘要:
      Objective To establish an HPLC method for concentration determination of lamotrigine in human serum,and to apply such method to the clinical work.Methods Serum samples were extracted by dichloromethane.Estazolam was taken as the internalstandard.Inertsil ODS?3 chromatographic column(4.6 mm×250 mm,5 μm)was used.The mobile phase was prepared with methanol∶ water=75∶25 at a flow rate of 1.0 mL/min.The detection wavelength was set at 225 nm with a column temperature of 25 ℃ and asample quantity of 10 μL.Results The linear regression equation of lamotrigine was Y=9.934 4X+0.808 3,r=0.999 8(n=7). The serum concentrations of lamotrigine showed a good linear relationship within the range of 1.00?18.00 μg/mL.Intraday precision, interday precision and stability all met the relevant requirements(Relative standard deviation being less than 15%).The sample re? covery and extraction recovery of 3.0 μg/mL,9.0 μg/mL and 18.0 μg/mL lamotrigine were all greater than 96% and 72% respec? tively.Conclusion The method is simple,fast and accurate,which can be used for therapeutic drug monitoring of lamotrigine.Therecommended plasma concentration of lamotrigine was 3?14 μg/mL which provides reference for clinically individualized and ratio?nal use of drugs.
