      目的探讨隐丹参酮对舌鳞状细胞癌细胞增殖、凋亡的影响及其作用机制。方法实验设置不同浓度隐丹参酮( CT)组、空白对照( NC)组、 pcDNA?con组、 pcDNA?GPR55组、 CT+si?con组、 CT+si?GPR55组。 MTT法检测细胞增殖抑制率;流式细胞术检测细胞凋亡; qRT?PCR检测 GPR55 mRNA的表达水平;蛋白质印迹法检测蛋白表达。结果 MTT法检测显示, 0 CT组、 2 CT组、 4 CT组、 8 CT组、 16 CT组、 32 CT组细胞增殖抑制率分别为( 0.00±0.02)%、(25.04±2.50)%、(68.56±5.86)%、(85.44±8.54)%、(88.25±8.82)%、(80.11±8.01)%,与不含隐丹参酮的细胞相比,不同浓度隐丹参酮处理组 TSCC细胞的增殖抑制率均显著升高( F=284.028,P<0.05)。隐丹参酮可抑制舌鳞状癌细胞增殖,促进细胞凋亡,抑制 Bcl?2、β?catenin表达,促进 Bax、 GPR55表达。过表达 GPR55也抑制舌鳞状癌细胞增殖,促进细胞凋亡,抑制 CyclinD1、Bcl?2表达,促进 Bax表达。低表达 GPR55逆转了隐丹参酮对舌鳞状癌细胞的增殖抑制、凋亡促进及对 β?catenin表达的抑制作用。与 NC组( 0.75±0.08)相比, CT组 TSCC细胞中 β?catenin表达水平( 0.26±0.03)显著降低;与 CT+si?con组( 0.30±0.03)相比, CT+si?GPR55组 TSCC细胞中 β?catenin表达水平( 0.62±0.06)显著升高( F=176.212,P<0.05)。结论隐丹参酮可抑制舌鳞状癌细胞的增殖,促进细胞凋亡,其机制可能与调控 GPR55及 Wnt/β?catenin信号通路有关。

      Objective To investigate the effect of cryptotanshinone on proliferation and apoptosis of tongue squamous cell carcino?ma cells and its mechanism.Methods Different concentrations of cryptotanshinone(CT)group,blank control(NC)group,pcDNA?con group,pcDNA?GPR55 group,CT+si?con group and CT+si?GPR55 group were set.MTT assay was used to detect cell prolifera? tion inhibition rate;flow cytometry was used to detect apoptosis;qRT?PCR was used to detect expression of GPR55 mRNA;West? ern Blot was used to detect protein expression.Results Cryptotanshinone inhibited the proliferation of tongue squamous carcinoma cells,promoted apoptosis,inhibited the expression of Bcl?2 and β?catenin,and promoted the expression of Bax and GPR55.Overex?pression of GPR55 also inhibits the proliferation of tongue squamous carcinoma cells,promotes apoptosis,inhibits the expression of CyclinD1 and Bcl?2,and promotes the expression of Bax.Low expression of GPR55 reversed the inhibitory effect of cryptotanshi?none on proliferation inhibition,apoptosis promotion and inhibition of β?catenin expression in tongue squamous carcinoma cells. Conclusion Cryptotanshinone can inhibit the proliferation of tongue squamous carcinoma cells and promote cell apoptosis.Themechanism may be related to the regulation of GPR55 and Wnt/β?catenin signaling pathways.
