      目的检测人源性多肽( DCD)在乳腺癌癌组织中的表达情况,研究 DCD的表达与病例资料之间的关系,初步讨论 DCD作为乳腺癌诊断生物学标志物的可能性,为乳腺癌的早期筛查及诊断提供新的方式。方法收集 2018年 6月至 2019年 1月于广州中医药大学附属广州中医院乳腺外科行乳腺癌手术 48例切除标本及其癌周正常组织标本,分为两组:实验组(癌组织)和对照组(正常组织),两组分别进行蛋白质印迹法( WB)检测及免疫组织化学( IHC)染色,比较癌组织和正常组织中 DCD的表达情况,以及研究其表达与病例资料之间的联系。结果在 WB检测中,与乳腺癌的正常组织相比, DCD在癌组织中的表达较高[(0.886±0.814)比( 0.641±0.482)t=5.330,P<0.001];在 IHC染色中, DCD在癌组织中 35例高表达, 13例低表达。而在对照组 48例正常组织中均为低表达,两组,差异有统计学意义( χ2=55.082,P<0.001); DCD表达情况与病例资料(年龄、绝经、临床分期、肿瘤长径、淋巴结转移、病理类型、分化程度等)之间无明显关系( P>0.05)。结论与乳腺正常组织相比,癌组织中的 DCD表达明显上调,其有望成为乳腺癌临床诊断的生物学标志物。

      Objective To investigate the relationship between the expression of Dermcidin(DCD)and the case data in patients with breast cancer by detecting the expression of DCD in cancer tissues,and to discuss the possibility of DCD as a biological mark?er for breast cancer diagnosis and to provide new ways for early screening and diagnosis of breast cancer.Methods Forty?eight cas?es of breast cancer resection specimens and their peri?normal tissues were collected from the Department of Breast Surgery,Guang?zhou Hospital of Traditional Chinese Medicine Affiliated to Guangzhou University of Chinese Medicine from June 2018 to January2019 and divided into two groups:experimental group(cancer tissue)and control group(normal tissue).Western blot(WB)and immunohistochemical(IHC)staining were conducted to compare DCD expression in cancer tissues and normal tissues.The relationbetween DCD expression and clinical data was studied.Results In WB detection,the expression of DCD in cancer tissues was higher than that in normal tissues[( 0.886±0.814)vs.(0.641±0.482),t=5.330,P<0.001]; In IHC staining,DCD was highly ex? pressed in 35 cases and low in 13 cases.However,the expression was low in 48 normal tissues of the control group,and the differ? ence between the two groups was statistically significant(χ2=55.082,P<0.001); There was no significant correlation between DCD expression and case data(age,menopause,clinical stage,tumor diameter,lymph node metastasis,pathological type,differentia? tion degree,etc.)( P>0.05).Conclusion Compared with normal breast tissue,the expression of DCD in cancer tissues is signifi? cantly up?regulated,which is expected to be a biomarker for clinical diagnosis of breast cancer.
