      目的探讨槲皮素对胶原诱导性关节炎小鼠Toll样受体4/核因子-κB(TLR4/NF-κB)信号通路的影响,阐明槲皮素治疗胶原诱导性关节炎的可能机制。方法将小鼠采用随机数字表法分为对照组(C组)、模型组(M组)、蜂毒肽组(Q组)和甲氨蝶呤组(MTX组),每组10只。采用Ⅱ胶原建立胶原诱导关节炎动物模型,Q组小鼠腹腔注射槲皮素(50 mg/kg),MTX组小鼠腹腔注射甲氨蝶呤(0.5 mg/kg),每3天1次,共3周。视觉模拟评分法进行关节炎评分。苏木精-伊红(HE)染色观察踝关节病理形态学变化。蛋白质印迹法(Western blotting)和荧光定量逆转录-聚合酶链反应(RT-PCR)测定软骨组织中TLR4、肿瘤坏死因子受体相关因子6(TRAF6)、髓样分化因子88(MYD88)、NF-κB p65蛋白和信使核糖核酸(mRNA)水平。结果与C组比较,M组足掌肿胀厚度升高[(4.85±0.64)mm比(2.16±0.53),P<0.05],软骨组织中TLR4、TRAF6、MYD88、NF-κBp65蛋白和mRNA水平升高(P<0.05);与M组比较,Q组(3.12±0.63)和MTX组(2.87±0.57)足掌肿胀厚度降低(P<0.05),病理评分(骨破坏评分、血管翳评分、滑膜炎评分及总分)明显降低(P<0.05),关节炎评分明显降低(P<0.05),软骨组织中TLR4、TRAF6、MYD88、NF-κBp65蛋白和mRNA水平降低(P<0.05);Q组和MTX组各指标差异无统计学意义(P>0.05)。结论槲皮素可抑制胶原诱导性关节炎小鼠关节炎症反应,其机制可能与槲皮素可抑制TLR2/NF-κB信号通路有关。

      Objective To investigate the effect of quercetin on Toll-like receptor 4/nuclear transcription factor- κB (TLR4/NF- κB) signaling pathway in collagen-induced arthritis mice, and to elucidate the possible mechanism of quercetin in the treatment of collageninduced arthritis.Methods Mice were assigned into control group (C group), model group (M group), melittin group (Q group) and methotrexate group (MTX group), with 10 mice in each group. Collagen Ⅱ was used to establish the collagen-induced arthritis animal model. The mice in Q group were injected intraperitoneally with quercetin (50 mg/kg); the mice in MTX group were intraperitoneally in-jected with methotrexate (0.5 mg/kg), once every three days, for 3 weeks. Visual analog scoring was used to perform an arthritis score.Hematoxylin-eosin (HE) staining was used to observe the pathological changes of the ankle joint. Western blot and fluorescence quanti-tative reverse transcription-polymerase chain reaction (RT-PCR) were used to determine the TLR4, tumor necrosis factor receptor-relat-pared with C group, the swelling thickness of foot palm was increased in group M [(4.85±0.64) mm vs. (2.16±0.53), P<0.05], the levels of TLR4, TRAF6, MYD88, NF-κB p65 protein and mRNA in cartilage tissue were increased (P<0.05) in group M. Compared with M group, the swelling thickness of the paws was decreased in the Q group (3.12±0.63) and the MTX group (2.87±0.57) (P<0.05), the path-ological scores (bone destruction score, vasospasm score, synovitis score and total score) were lower (P<0.05), the arthritis score were significantly lower (P<0.05), the levels of TLR4, TRAF6, MYD88, NF-κBp65 protein and mRNA in cartilage tissues were decreased in the Q group and the MTX group (P<0.05). There were no significant differences in each indexs between the Q group and the MTX group(P>0.05).Conclusion Quercetin can inhibit joint inflammation in mice with collagen-induced arthritis, and its mechanism may be re-lated to the inhibition of TLR2/NF-κB signaling pathway by quercetin.
