      目的研究蜂斗菜素对宫颈癌细胞增殖、细胞周期和凋亡的影响及潜在机制。方法以蜂斗菜素0 μmol/L为对照,用5、15、45 μmol/L的蜂斗菜素处理宫颈癌SiHa细胞,四甲基偶氮唑盐比色法(MTT)检测细胞在24、48和72 h的增殖活性,蛋白质印迹法(Western blotting)检测SiHa细胞中细胞周期蛋白D1(cyclin D1)、细胞周期蛋白依赖性激酶2(CDK2)、活化的半胱氨酸天冬氨酸蛋白酶3(Cleaved-caspase-3)、活化的半胱氨酸天冬氨酸蛋白酶9(Cleaved-caspase-9)、磷脂酰肌醇3-激酶(PI3K)、p-PI3K、丝/苏氨酸蛋白激酶(AKT)和磷酸化的丝/苏氨酸蛋白激酶(p-AKT)蛋白的表达,流式细胞术检测细胞周期和凋亡率;然后用AKT信号通路激活剂IGF-1处理细胞,检测其对蜂斗菜素处理后的SiHa细胞的影响。结果与对照相比,5、15、45 μmol/L的蜂斗菜素处理宫颈癌SiHa细胞后,在24、48和72 h细胞OD值均显著降低,且呈浓度和时间依赖性(P<0.05),细胞中cyclinD1、CDK2、p-PI3K和p-AKT表达量均降低(P<0.05),Cleaved caspase-3和Cleaved caspase-9蛋白表达升高,细胞凋亡率升高,G0~G1期细胞百分比显著升高(P<0.05),S期细胞百分比显著降低(P<0.05),说明蜂斗菜素可使SiHa细胞滞留在G0~G1期,抑制AKT信号通路,抑制细胞增殖,促进细胞凋亡;AKT信号通路激活剂IGF-1部分逆转了蜂斗菜素对SiHa细胞增殖、凋亡、细胞周期和AKT信号通路的影响。结论蜂斗菜素可能通过AKT信号通路调控宫颈癌细胞增殖、细胞周期和凋亡。蜂斗菜素对宫颈癌具有潜在抑制作用。

      Objective To investigate the effects and the underlying mechanism of petasin on the cell cycle, proliferation and apopto-sis of cervical cancer cells.Methods Taking petasin 0 μmol/L as control, cervical cancer SiHa cells were treated with 5, 15, and 45 μmol/L of petasin. The proliferation activity of SiHa cells at 24 h, 48 h and 72 h was detected by MTT assay. Expression levels of cyclin D1, CDK2, Cleaved caspase-3, Cleaved caspase-9, p-PI3k, PI3K, p-AKT and AKT in cervical cancer cell lines were detected by West-ern blot. Cell cycle and apoptosis rates were detected by flow cytometry. The cells were then treated with IGF-1, an activator of the AKT signaling pathway, to detect its effect on SiHa cells treated with petasin.Results Compared with the control, the cell OD values of cer-vical cancer SiHa cells treated with 5, 15 and 45 μmol/L petasin were significantly decreased at 24 h, 48 h and 72 h in a concentration and time dependent manner (P<0.05), and the expression levels of cyclin D1, CDK2, p-PI3K and p-AKT in the cells were all decreased(P<0.05), the levels of Cleaved caspase-3 and Cleaved caspase-9 protein were increased, the apoptosis rate was increased, cell percent-age of G0-G1 phase was increased significantly (P<0.05), cell percentage of S phase was decreased significantly (P<0.05), indicating that petasin can make SiHa cells stay in G0-G1 phase, inhibit AKT signaling pathway, inhibit cell proliferation and promote cell apopto-sis. IGF-1, an activator of AKT signaling pathway, partially reversed the effects of petasin on proliferation, apoptosis, cell cycle and AKT signaling pathway of SiHa cells.Conclusions Petasin may regulate proliferation, cell cycle and apoptosis of cervical cancer cells through AKT signaling pathway. Petasin has potential inhibitory effect on cervical cancer.
