      目的探讨止麻消痰活血汤对老年心肌梗死大鼠神经调节蛋白 -1(NRG-1)/受体酪氨酸蛋白激酶 erbB3(ErbB3)通路及心室重构的影响。方法研究起止时间为 2019年 2—10月,大鼠来源于上海邦耀生物公司。制备老年心肌梗死模型大鼠,以随机数字表法分为模型组、止麻消痰活血汤低中高剂量组( 10 mL/kg、20 mL/kg、30 mL/kg)、地尔硫.组( 2.61 mg/kg),每组 12只,另取 12只 SD老年大鼠设为假手术组,分组处理后,测定各组大鼠左心室质量指数;苏木精 -伊红( HE)、马松( Mosson)染色分别检测大鼠心肌病理形态变化及心室纤维化程度;酶联免疫吸附测定(ELISA)检测大鼠血清转化生长因子 -β1(TGF-β1)、白细胞介素 -6(IL-6)、肿瘤坏死因子 -α(TNF-α)水平;蛋白质印迹法检测大鼠心肌 NRG-1/ErbB3通路相关蛋白的表达。结果与假手术组比较,模型组大鼠心肌纤维扭曲、断裂,心肌细胞肥大、坏死,可见炎性细胞浸润等病理损伤,且胶原纤维显著增多,纤维化严重,血清 TGF-β1、IL-6及 TNF-α水平、左心室质量指数[( 23.27±1.84).比( 12.23±0.85).]显著升高( P<0.05),心肌 NRG-1/ErbB3通路相关蛋白 NRG-1[( 0.30±0.04)比( 2.16±0.37)]、 p-ErbB3/ErbB3[( 0.09±0.02)比( 1.02±0.20)]显著降低( P<

      Objective To investigate the effect of Zhima Xiaotan Huoxue decoction on the neuregulin-1/receptor tyrosine-protein ki-nase erbB3 (NRG-1/ErbB3) pathway and ventricular remodeling in aged myocardial infarction rats.Methods The study started andended from February to October 2019, and the rats were sourced from Shanghai Bangyao Biological Company. The aged myocardial in-farction model rats were prepared and randomly divided into the model group, Zhima Xiaotan Huoxue decoction low (10 mL/kg), medi-um (20 mL/kg) and high (30 mL/kg) dose groups, and diltiazem (2.61 mg/kg) group, with 12 rats in each group, and another 12 Sprague-Dawley (SD) rats were selected as the sham operation group. The left ventricular mass index of the rats in each group was measured. He-matoxylin-eosin (HE) staining and Masson staining were used to detect the pathological changes in the myocardium and the degree ofventricular fibrosis. The levels of serum transforming growth factor-β1 (TGF-β1), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) were detected by enzyme-linked immunosorbent assay (ELISA). The expression of NRG-1/ErbB3 pathway-related proteins in the rat myocardium was detected by Western blotting.Results Compared with the sham operation group, the myocardial fibers of the modelgroup were twisted and broken, the myocardial cells were hypertrophied and necrotic, inflammatory cell infiltration and other pathologi-cal damage were observed, collagen fibers were significantly increased, and fibrosis was severe. Serum levels of TGF-β1, IL-6 and TNF-α and the left ventricular mass index [(23.27±1.84). vs. (12.23±0.85).] were significantly increased (P < 0.05). The myocardial NRG-1/ErbB3 pathway-related proteins NRG-1 [(0.30±0.04) vs. (2.16±0.37)] and p-ErbB3/ErbB3 [(0.09±0.02) vs. (1.02±0.20)] were signifi-cantly decreased (P < 0.05). Compared with the diltiazem group, the pathological damage and fibrosis symptoms of the Zhima Xiaotan Huoxue decoction low-, medium-and high-dose groups and the thiazolone group were improved, and the levels of serum TGF-β1, IL-6 and TNF-α and the left ventricular mass index [(19.92±1.47)., (16.36±1.06)., (12.89±0.87)., (12.84±0.86). vs. (23.27±1.84).] were lower (P < 0.05). The myocardial NRG-1/ErbB3 pathway-related proteins NRG-1 [(0.94±0.18), (1.27±0.23), (2.09±0.31), (2.11±0.33) vs. (0.30±0.04)] and p-ErbB3/ErbB3 [(0.27±0.04), (0.51±0.08), (0.92±0.16), (0.94±0.18) vs. (0.09±0.02)] were higher (P < 0.05), and the Zhima Xiaotan Huoxue decoction groups were dose-dependent (P < 0.05). Compared with the diltiazem group, there was no sig-nificant difference in each index between the high-dose Zhima Xiaotan Huoxue decoction group (P > 0.05).Conclusion Zhima Xiao. tan Huoxue decoction can activate the NRG-1/ErbB3 pathway, reduce inflammation in rats with myocardial infarction, relieve the patho-logical injury and fibrosis of myocardial tissue, and improve the symptoms of ventricular remodeling in rats.
