      目的:研究长链非编码RNA PCGEM1(lncRNA PCGEM1)和信号传导转录激活因子3(STAT3)在卵巢癌中的表达关系及意义。方法:选取 2013年3月-2014年12月本院收诊卵巢癌患者108例,卵巢摘除手术中留取卵巢组织,另因卵巢囊肿需作手术切除但已证实无肿瘤细胞的正常卵巢组织108例,采用实时定量PCR(RT-qPCR)检测卵巢癌及正常卵巢组织中lncRNA PCGEM1、STAT3 mRNA 表达水平,采用免疫组化法检测STAT3磷酸化蛋白阳性表达率,采用Pearson法分析卵巢癌组织中lncRNA PCGEM1、STAT3 mRNA 表达水平相关性,采用Kaplan-Meier生存曲线分析lncRNA PCGEM1、STAT3 磷酸化蛋白表达与卵巢癌患者5年生存率的关系;采用Cox比例风险模型分析影响卵巢癌患者预后的危险因素。结果:与正常卵巢组织比较,卵巢癌组织lncRNA PCGEM1、STAT3 mRNA表达水平、STAT3蛋白阳性表达率显著升高(P<0.05);卵巢癌组织lncRNA PCGEM1、STAT3蛋白阳性表达与患者年龄无关(P>0.05),与FIGO分期、病理分级、铂类耐药及淋巴结转移有关(P<0.05)。卵巢癌组织lncRNA PCGEM1、STAT3 mRNA表达水平呈正相关(P<0.05)。lncRNA PCGEM1高表达组5年生存率分别为8.33%,显著低于低表达组54.35%(c2=39.51,P<0.001),STAT3蛋白阳性表达组5年生存率为13.16%,显著低于阴性表达组68.75%(c2=26.16,P<0.001)。lncRNA PCGEM1高表达、STAT3蛋白阳性表达、顺铂耐药及淋巴结转移是影响卵巢癌患者预后的独立危险因素(P均<0.05)。结论:卵巢癌组织中lncRNA PCGEM1、STAT3均高表达,且二者呈正相关,与卵巢癌患者FIGO分期、病理分级、铂类耐药、淋巴结转移及5年生存率有关,是影响卵巢癌患者预后的独立危险因素,有潜力成为监测卵巢癌病情发展及评估预后的生物标志。

      Objective: To study the expressions and significances of long non-coding RNA PCGEM1 (lncRNA PCGEM1) and signal transduction activator 3 (STAT3) in ovarian cancer. Methods: 108 patients with ovarian cancer admitted to our hospital from March 2013 to December 2014 were selected, and ovarian tissues were collected during ovariectomy. In addition, 108 cases of normal ovarian tissues with ovarian cysts which need to be removed surgically but without tumor cells were selected. The expression levels of lncRNA PCGEM1 and STAT3 in ovarian cancer and normal ovarian tissues were detected by real-time quantitative PCR (RT-qPCR), the positive expression rate of STAT3 phosphorylated protein was detected by immunohistochemistry, Pearson method was used to analyze the correlation between the expression levels of lncRNA PCGEM1 and STAT3 in ovarian cancer tissues, Kaplan-Meier survival curve was used to analyze the relationships between the expressions of phosphorylated proteins of lncRNA PCGEM1 and STAT3 and the 5-year survival rate of ovarian cancer patients, and Cox proportional hazard model was used to analyze the risk factors affecting the prognosis of ovarian cancer patients. Results: Compared with normal ovarian tissues, the expression levels of lncRNA PCGEM1, STAT3 mRNAs and the positive rate of STAT3 protein in ovarian cancer tissues were significantly increased (P < 0.05). The positive expressions of lncRNA PCGEM1 and STAT3 proteins in ovarian cancer tissues were not related to age (P > 0.05), but related to FIGO stage, pathological grade, platinum resistance and lymph node metastasis (P < 0.05). The expression levels of lncRNA PCGEM1 and STAT3 in ovarian cancer tissues were positively correlated (P < 0.05). The 5-year survival rate of the high expression group of lncRNA PCGEM1 was 8.33%, which was significantly lower than 54.35% of the low expression group (c2 = 39.51, P < 0.001), and the 5-year survival rate of the STAT3 positive expression group was 13.16%, which was significantly lower than 68.75% of the negative expression group (c2 = 26.16, P < 0.001). High expression of lncRNA PCGEM1, positive expression of STAT3 protein, cisplatin resistance and lymph node metastasis were independent risk factors affecting the prognosis of ovarian cancer patients (all P < 0.05). Conclusions: The lncRNA PCGEM1 and STAT3 are highly expressed in ovarian cancer tissues, they are positively correlated, and are positively correlated with FIGO stage, pathological grade, platinum resistance, lymph node metastasis and 5-year survival rate of ovarian cancer patients. They are independent risk factors affecting the prognosis of ovarian cancer patients, and have potential to be used as biomarkers to monitor the development of ovarian cancer and evaluate the prognosis.
