      目的研究西格列汀对高糖诱导内皮细胞凋亡的改善作用及机制。方法培养人脐静脉内皮细胞(HUVECs)并分组,对照组用含有 5.5 mmol/L葡萄糖的 DMEM处理,高糖组用含有 33.3 mmol/L葡萄糖的 DMEM处理,西格列汀组用含有 33.3 mmol/L葡萄糖及 20 μmol/L西格列汀的 DMEM处理,西格列汀 +LY组用含有 33.3 mmol/L葡萄糖、 20 μmol/L西格列汀、 10 μmol/L LY294002的 DMEM处理。检测各组细胞凋亡率及细胞中凋亡基因、人磷酸化磷酸肌醇 3激酶(p?PI3K)、磷酸化蛋白激酶 B(p?AKT)的表达量。结果高糖组的细胞凋亡率及 Cleaved?caspase?9[0.91(0.71,1.12)比 0.43(0.34,0.59)]、 Cleaved?cas? pase?3[0.75(0.67,0.93)比 0.45(0.36,0.58)]的蛋白表达量明显高于对照组, B细胞淋巴瘤 /白血病 ?2(Bcl?2)[0.23(0.17,0.36)比 0.60(0.45,0.72)]、 p?PI3K[0.40(0.22,0.61)比 0.94(0.78,1.21)]、 p?AKT[0.27(0.13,0.34)比 0.66(0.40,0.79)]的蛋白表达量明显低于对照组;西格列汀组的细胞凋亡率及 Cleaved?caspase?9[0.32(0.26,0.43)比 0.91(0.71,1.12)]、 Cleaved?caspase?3[0.34(0.24,0.48)比 0.75(0.67,0.93)]的蛋白表达量明显低于高糖组, Bcl?2[0.69(0.58,0.83)比 0.23(0.17,0.36)]、 p?PI3K[0.87(0.67,0.99)比 0.40(0.22,0.61)]、 p?AKT[0.65(0.48,0.73)比 0.27(0.13,0.34)]的蛋白表达量明显高于高糖组;西格列汀 +LY组的细胞凋亡率及 Cleaved?caspase?9[0.72(0.58,0.86)比 0.32(0.26,0.43)]、 Cleaved?caspase?3[0.68(0.54,0.73)比 0.34(0.24,0.48)]的蛋白表达量明显高于西格列汀组, Bcl?2[0.37(0.29,0.45)比 0.69(0.58,0.83)]、 p?PI3K[0.49(0.34,0.58)比 0.87(0.67,0.99)]、 p?AKT[0.32(0.24,0.38)比 0.65(0.48,0.73)]的蛋白表达量明显低于西格列汀组。结论西格列汀能够抑制高糖诱导的内皮细胞凋亡且该作用与激活 PI3K/AKT通路有关。

      Objective To study the improvement effect and mechanism of sitagliptin on high glucose induced apoptosis of endothe? lial cells.Methods Human umbilical vein endothelial cells(HUVECs)were cultured and grouped.The control group was treated with DMEM containing 5.5 mmol/L glucose,the high glucose group was treated with DMEM containing 33.3 mmol/L glucose,the si? tagliptin group was treated with DMEM containing 33.3 mmol/L glucose and 20 μmol/L sitagliptin,and the sitagliptin+LY group was treated with DMEM containing 33.3 mmol/L glucose,20 μmol/L sitagliptin and 10 μmol/L LY294002.The apoptotic rate and the expression of apoptotic genes,p?PI3K and p?AKT were detected.Results The apoptotic rate and the expression of Cleaved?cas? pase?9[0.91(0.71,1.12)vs. 0.43(0.34,0.59)]and Cleaved?caspase?3[0.75(0.67,0.93)vs. 0.45(0.36,0.58)]in high glucose group were significantly higher than those in control group,while the expression of Bcl?2[0.23(0.17,0.36)vs. 0.60(0.45,0.72)], p?PI3K[0.40(0.22,0.61)vs. 0.94(0.78,1.21)] and p?AKT[0.27(0.13,0.34)vs. 0.66(0.40,0.79)] were significantly lower than those in control group.The apoptotic rate and the expression of Cleaved?caspase?9[0.32(0.26,0.43)vs. 0.91(0.71,1.12)] and Cleaved?caspase?3[0.34(0.24,0.48)vs. 0.75(0.67,0.93)]in sitagliptin group were significantly lower than those in high glu? cose group,and the expression of Bcl?2[0.69(0.58,0.83)vs. 0.23(0.17,0.36)],p?PI3K[0.87(0.67,0.99)vs. 0.40(0.22, 0.61)]and p?AKT[0.65(0.48,0.73)vs. 0.27(0.13,0.34)]were significantly lower than those in control group.The apoptotic rate and the expression of Cleaved?caspase?9[0.72(0.58,0.86)vs. 0.32(0.26,0.43)] and Cleaved?caspase?3[0.68(0.54,0.73)vs. 0.34(0.24,0.48)] in Sitagliptin+LY group were significantly higher than those in Sitagliptin group,while the expression levels of Bcl?2[0.37(0.29,0.45)vs. 0.69(0.58,0.83)],p?PI3K[0.49(0.34,0.58)vs. 0.87(0.67,0.99)]and p?AKT[0.32(0.24,0.38)vs. 0.65(0.48,0.73)] were significantly lower than those in Sitagliptin group.Conclusion Sitagliptin can inhibit high glucose in? duced apoptosis of endothelial cells,which is related to the activation of PI3K/AKT pathway.
