目的 建立同时测定三叶青药材中胡萝卜苷和β-谷甾醇含量的方法。方法 采用HPLC-ELSD法,岛津VPN-ODS C18柱(150 mm×4.6 mm,5 μm)为色谱柱,以甲醇为流动相,体积流量为0.7 mL·min-1,柱温为30℃;漂移管温度60℃,氮气体积流量1.8 L ·min-1。结果 胡萝卜苷和β-谷甾醇分别在0.1~2.0 μg和0.2~4.0 μg范围内呈良好线性关系,回归方程分别为Y= 0.696X + 2.737,r=0.999 7;Y= 0.752 X + 2.909,r=0.999 6。平均回收率分别为99.32%、100.13%。结论 该方法简便,结果准确、可靠,可作为三叶青药材中胡萝卜苷和β-谷甾醇的含量测定方法,为三叶青质量控制提供依据。 |

Objective To develop a method for simultaneous determination of daucosterol and β-sitosterol in Tetrastigmatis hemsleyani Diels et. Gilg. Methods HPLC-ELSD method was used to determine daucosterol and β-sitosterol. The separation was performed on a Shim-pack VPN-ODS C18 column(150 mm×4.6 mm,5 μm)with methanol as the mobile phase at the flow rate of 0.7ml.min-1, column temperature of 30℃. The temperature in drift tube was 60℃ and nitrogen flow rate was 1.8 L.min-1. Results: The linear ranges of daucosterol was 0.1~2.0 μg, the regression equation was Y = 0.696X + 2.737(r=0.999 7), and the average recovery rate was 99.32%. The linear ranges of β-sitosterol was 0.2~4.0 μg, the regression equation was yY= 0.752 X + 2.909(r=0.999 6), and the average recovery rate was 100.13%. Conclusion The method is simple, accurate and reliable, which can be used as quantitative determination method for daucosterol and β-sitosterol in Tetrastigmatis hemsleyani Diels et. Gilg. |
