目的 研究辐射对人胶质瘤U251细胞COX-2和survivin蛋白表达的影响。方法 体外培养人胶质瘤U251细胞,随机分为实验组和对照组,实验组经过10 Gy剂量X线照射,对照组无特殊处理,培养48 h后用免疫组化法分别检测两组细胞中COX-2和survivin蛋白的表达情况。结果 COX-2和survivin蛋白在人胶质瘤U251细胞的细胞质和细胞核中表达,X线照射48 h后检测出COX-2和survivin蛋白在细胞内的表达较对照组均有明显增加(P<0.05)。结论 辐射可诱导胶质瘤细胞COX-2和survivin蛋白表达的增加,这可能是胶质瘤存在放射抵抗的机制之一。 |

Objective To observe the effect of radiation on the expression of COX-2 and survivin proteins in human glioma cell line U251.Methods The human glioma cell line U251 was divided into control group(0 Gy) and experimental group(10 Gy).We detected the expression of COX-2 and survivin proteins in cells of two groups by immunohistochemical method, and observed the change of expression induced by radiation. Results The positive stainings of COX-2 and survivin proteins were mainly located in cytoplasm and nucleus of U251 cells. The expression of COX-2 and survivin in U251 cells after radiation were significantly higher than the expression without radiation (P<0.05).Conclusion The expression of COX-2 and survivin proteins are increased induced by radiation, which may become one of mechanisms about the radioresistance of glioma cells. |
