      目的 探索青蒿琥酯(ART)对人颚部小涎腺腺样囊性癌(NACC)细胞增殖、侵袭能力的影响。方法 用MTT法检测经37.5、150、600 μmol·L-1ART干预24、48 h后,NACC细胞体外增殖水平的变化;Transwell法检测37.5、150、600 μmol·L-1 ART干预48 h后,NACC细胞体外侵袭能力的变化;Western Blot法检测ART干预48 h后,NACC细胞中转录因子SP1蛋白表达水平的变化;各实验方法均设立空白对照组及5 μmol·L-1顺铂(DDP)阳性对照组。结果 与对照组比较,ART作用于NACC细胞24 h时即表现出对其增殖能力的抑制作用(F=12.071,P<0.001),作用时间延长至48 h时,抑制作用更加明显(F =169.098,P<0.001);Transwell实验中,与对照组相比,37.5、150、600 μmol·L-1 ART组穿膜细胞数量明显减少(F=139.939,P<0.001);Western Blot实验结果显示:与空白对照组相比,37.5、150、600 μmol·L-1 ART及5 μmol·L-1 DDP阳性对照组中SP1蛋白表达水平均出现不同程度的下降(F=200.998,P<0.001)。结论 青蒿琥酯可以有效抑制NACC细胞体外增殖及侵袭能力,并下调转录因子SP1表达,二者间可能存在联系。

      Objective To explore the effect of artesunate (ART) on proliferation and invasion of novel human palatal salivary gland cell line of adenoid cystic carcinoma (NACC). Methods The changes of in vitro proliferation of NACC cells after being intervened for 24 and 48 hours by 37.5,0,600 μmol·L-1 ART were analyzed by using MTT assay; the anti-invasion effect of ART at 37.5,0,600 μmol·L-1 were tested by Transwell assay; the expression level of SP1 protein in each group was detected by Western Blot method. All experiments were performed with a control group and a positive control group with 5 μmol·L-1 DDP.Results Compared with the control group, the effect of ART on the proliferation of NACC cells was significantly increased after treated for 24 hours (F=12.071, P<0.001), and the inhibition was more obvious than that by extending the intervention time to 48 hours (F=169.098, P<0.001). In Transwell assay, the number of cells passing through the membrane in experimental group (37.5,0,600 μmol·L-1 ART groups) was significantly decreased, compared with the control group (F=139.939,P<0.001).Western Blot results showed that the expression levels of SP1 protein in 37.5,0,600 μmol·L-1 ART and 5 μmol·L-1 DDP positive control group were decreased (F=200.998, P<0.001), compared with the blank control group. Conclusions Artesunate could effectively inhibit the NACC cell proliferation and invasive ability, and down-regulate the expression of transcription factor SP1.
