目的 筛选适合用于分离纯化东风桔总生物碱的树脂。方法 采用酸性染料比色法测定总生物碱的含量,以静态吸附率及解吸率为指标,对15种不同类型的树脂进行筛选,然后通过单因素法考察上样液pH值、吸附温度、吸附时间等对树脂吸附效果进行优化。结果 001*7阳离子交换树脂对东风桔总生物碱效果较好,总生物碱的吸附率达到73.51%,解吸率达到90.58%,在单因素试验中,上样液pH=2~3,T=50℃,吸附6 h时,具有最优的吸附效果。结论 001*7阳离子交换树脂适合用于东风桔总生物碱的分离纯化,为其进一步的富集工艺研究及新药研发提供实验参考。 |

Objective To select proper resins for isolation and purification of total alkaloids from Atalantia buxifolia. Methods The content of total alkaloids was determined by acid dye-colorimetry. With the adsorption rate and desorption rate of total alkaloids as the observation indexes, the optimized resin was selected by screening 15 types of resins. And then the resin adsorption influence factors were investigated by single-factor experiments based on pH, temperature and adsorption time. Results The 001*7 cation-exchanging resin had good adsorption and desorption activities, with an adsorption rate being 73.51% and a desorption rate being 90.58%. The optimum adsorption process:the extracting solution pH=2~3, T=50 ℃, and adsorbing time 6 hours. ConclusionThe 001*7 cation-exchanging resin could be used for total alkaloids enrichment, which could also provide a reference for further study. |
