目的 探讨武当地区马尾松松针中原花青素的含量测定方法。方法 采用高效液相色谱法测定松针中原花青素的含量。色谱柱为Sum Fortis C18(4.6 mm×150 mm,5 μm),流动相为乙腈∶水(15∶85),流速为0.6 mL·min-1,柱温30℃,检测波长为280 nm。结果 经方法学考察,原花青素在0.258~2.58 g·L-1范围内,峰面积与所试浓度线性关系良好,准确度高,精密度好。武当地区松针样品中原花青素含量为44.65 mg·g-1。结论 该方法操作简便,有效成分原花青素的含量较高,开发前景广阔,旨在其进一步研究提供依据。 |

Objective To discuss methods for the content determination of proanthocyandins in pine needles in Wudang area. Methods HPLC-DAD was used to determine proanthocyandin content of pine needles. The Column was Sum Fortis C18 (4.6 mm×150 mm, 5 μm) with the mobile phase of acetonitrile-water (15∶85). The flow rate was 0.6 mL·min-1. The temperature was 30℃, and the detective wavelength was 280 nm.Results The proanthocyandin of pine needles was well separated with other substances. The proanthocyandin’s peak area had a good linear relationship within the concentration range from 0.258 to 2.58 g·L-1. The results of methodological study demonstrated that the method met the requirements of the determination. The proanthocyandins contents of pine needles were 44.65 mg·g-1. Conclusions The method was simple and effective contents of procyanidins were high, thus providing basis for its further study with promising prospects. |
