      目的 通过选择10 nmol·L-1芬太尼作用于鼻咽癌细胞系CNE2和HONE1,研究芬太尼对鼻咽癌细胞迁移能力的影响。方法 取对数生长期的鼻咽癌细胞系CNE2和HONE1,选择10 nmol·L-1芬太尼分别作用于鼻咽癌细胞系30 min和1 h,Western blot检测p-Src和Src表达,Transwell检测细胞迁移能力;构建Src过表达质粒,转染鼻咽癌细胞系,观察鼻咽癌细胞形态,检测细胞的迁移能力。结果 10 nmol·L-1芬太尼处理鼻咽癌细胞CNE2和HONE1 48 h后,细胞呈现出类间质向上皮转化的形态特征,同时,与对照组相比,加入芬太尼处理组的鼻咽癌细胞其迁移能力显著降低;应用芬太尼分别处理鼻咽癌细胞30 min和1 h后,p-Src水平显著下调;此外,与转染空载体质粒的对照组相比,过表达Src的细胞迁移能力明显增强,而在过表达Src的细胞中同时联合芬太尼处理后,细胞的迁移能力没有被抑制。结论 芬太尼可能通过抑制Src通路活化抑制鼻咽癌细胞侵袭转移。

      Objective To investigate the effect of fentanyl on nasopharyngeal carcinoma cells(NPC) metastasis by treating NPC lines CNE2 and HONE1 with fentanyl (10 nmol·L-1) in this study.Methods Nasopharyngeal carcinoma cells (CNE2 and HONE1) in logarithmic growth phase were pretreated with fentanyl (10 nmol·L-1) for 30 min and 1 hour.Western blot was used to test the expression levels of p-Src and Src and Transwell test was used to evaluate cell metastatoc ability.Src over-expression plasmid was constructed and transfected into NPC cell lines and NPC cellular morphology and metastatic ability were observed.Results NPC cell lines (CNE2 and HONE1) exhibited mesenchymal-to-epithelial like transformation;furthermore,the metastatic ability of NPC cells decreased obviously compared with control group 48 hours after fentanyl treatment.The p-Src expression level was down-regulated after fentanyl treatment for 30 min and 1 hour.However,over-expressive Src enhanced the metastatic ability of NPC cells obviously,and in combination with fentanyl treatment over-expressive Src plasmid transfection did not inhibit the metastatic ability of NPC cells.Conclusions Fentanyl may inhibit nasopharyngeal carcinoma cells by inhibiting Src pathway invasion and metastasis.
