| 目的 建立高效液相法同时测定混合核苷片中腺嘌呤核苷、鸟嘌呤核苷、尿嘧啶核苷、胞嘧啶核苷含量的方法。方法 采用C18(250 mm×4.6 mm,5 μm)色谱柱；流动相:以0.01 mol·L-1磷酸二氢钾溶液为流动相A,甲醇为流动相B,梯度洗脱；流速:1.0 mL·min-1；检测波长:260 nm。用保留时间定性,以峰面积按外标法定量。结果 四种核苷分离良好,线性关系良好(r=0.999 6),平均加样回收率在98.4%～101.9%, RSD%≤2.7%。结论 该方法简便、准确,可用于混合核苷片中四种核苷的定性和定量分析。 |

| Objective To establish an HPLC method for the determination of four nucleosides in Mixed Nucleoside Tablets.Methods The separation was performed on C18 column(250 mm×4.6 mm,5 μm),eluted with 0.01 mol·L-1 potassium dinydrogen phosphate solution and methanol in gradient mode.The flow rate was 1.0 mL·min-1,and the detection wave length was 260 nm.With the retention time as for qualitative analysis,the peak area was quantified by the external standard method.Results Four nucleosides were separated well with good linear relation(r=0.999 6).The average recoveries were between 98.4% and 101.9%,and RSD was lower or equal to 2.7%.Conclusion The method was simple and accurate,so it can be used for the quantitative and qualitative detection of four nucleosides in Mixed Nucleoside Tablets. |
