目的 构建安全、稳定、免疫细胞靶向亚单位疫苗佐剂-传递系统。 方法 以磷脂酰丝氨酸为材料,采用乳化冻干法制备甘露糖修饰脂质卷,作为包封抗原载体。 结果 乳化冻干法制备脂质卷平均粒径为950 nm,呈杆状,对于模型抗原包封率为38%,能够保护抗原在室温条件下2周内结构完整,在37 ℃中性缓冲液中48 h抗原释放低于42%,并能够有效将所包封物质传递入免疫细胞。 结论 采用乳化-冻干法制备的脂质卷能够有效包封抗原,并提高其稳定性及免疫细胞靶向性,是一种有效的亚单位疫苗佐剂-传递系统。 |

Objective To establish a safe,stable and immunocyte-targeting subunit vaccine adjuvant-delivery system. Methods The mannose modified cochleates composed of phosphatidylserine (PS) were prepared through emulsification-lyophilization procedure for entrapping vaccine antigens. Results The prepared mannosylated cochleates (MC) had a rod-like shape,a size of around 950 nm and entrapment efficiencies for model antigen ovalbumin (OVA) of 38%.In vitro release test indicated that less than 42% of the associated OVA was liberated from the MC after 48 h incubation at 37 ℃ in a buffered neutral solution.Moreover,the MC maintained the antigens’ integrity for 2 weeks at room temperature and could efficiently deliver the loaded antigens to immunocytes. Conclusions The MC prepared by emulsification-lyophilization procedure can efficiently entrap antigens,enhancing subunit vaccine stability and delivery of antigens to immunocytes,and thus could be an effective vaccine adjuvant-delivery system (APC). |
