目的 建立高效液相色谱法(HPLC)和液质联用法(HPLC-MS)测定保健食品中非法添加降糖药物的定量及定性方法。方法 样品经甲醇涡旋离心提取后,采用HPLC和HPLC-MS测定。色谱条件:C18 柱(250 mm×4.6 mm,5 μm);流动相以甲醇-0.01 moL·L-1乙酸铵进行梯度洗脱;流速1.0 mL·min-1;检测波长:235 nm;柱温:30 ℃;检测器为二极管阵列检测器。质谱条件:以电喷雾电离子源(ESI+)采集数据。结果 非法添加降糖药物在各自测定范围内线性范围良好,平均回收率为92.51%~98.01%;质谱分辨率良好,专属性高。结论 该方法操作简便,灵敏度高,重现性好,专属性高。 |

Objective To establish an HPLC and HPLC-MS determination methods for illegal addition of hypoglycemic substances to health foods.Methods Samples were extracted by centrifugal vortexing methanol and determined by HPLC and HPLC-MS.The HPLC analysis was performed on a C18 column (250 mm×4.6 mm,5 μm) with methanol-0.01 moL·L-1 ammonium acetate as the mobile phase at a flow rate of 1.0 mL·min-1;the detection wavelength was at 235 nm;the column oven temperature was 30 ℃.The detector was DAD.The detector of the HPLC-MS is ESI+.Results The illegally added hypoglycemic substances had a good linear range,and the average recovery was 92.51% to 98.01%.HPLC-MS had a good separation and high specificity.Conclusions The method is simple,sensitive,reproducible,and highly specific for the determination of illegal addition of hypoglycemic substances to health foods. |
