目的 探究硼替佐米对伯基特淋巴瘤Raji细胞体外抑制作用及其可能机制。方法 MTT实验检测硼替佐米对伯基特淋巴瘤Raji细胞活力的影响;Annexin V-FITC /PI双染流式细胞仪检测硼替佐米对伯基特淋巴瘤Raji细胞凋亡的影响;Western blotting实验检测硼替佐米对伯基特淋巴瘤Raji细胞中cleaved caspase-3和p-NF-κB p65表达的影响。结果 硼替佐米浓度和时间依赖性抑制Raji细胞活力(P<0.05)和促进细胞凋亡(P<0.05);Western blotting实验显示硼替佐米浓度和时间依赖性抑制Raji细胞NF-κB p65表达并促进caspase-3的剪切(P<0.05)。结论 硼替佐米促进Raji细胞凋亡,其机制可能与抑制p-NF-κB p65和上调cleaved-caspase-3表达有关。 |

Objective To explore inhibitory effect of bortezomib on Burkitt lymphoma Raji cells and its possible mechanism. Methods The effect of bortezomib on Burkitt lymphoma Raji cell viability was determined by MTT assay. The effect of bortezomib on Burkitt lymphoma Raji cell apoptosis was determined by annexin V-FITC/PI staining using flow cytometer. The effect of bortezomib on the protein expressions of p-NF-κB p65 and cleaved-caspase-3 were detected by Western blotting. Results The data showed that bortezomib inhibited Raji cell viability (P<0.05) and promoted Burkitt lymphoma Raji cell apoptosis (P<0.05) were related in a dose-dependent manner and a time-dependent manner. Western blotting data indicated that the expression of p-NF-κB p65 was decreased and the expression of cleaved-caspase-3 was increased after bortezomib treatment in a dose dependent manner and a time-dependent manner(P<0.05). Conclusion Bortezomib can promote Raji cell apoptosis may via downregulating the expression of p-NF-κB p65 and upregulating the expression of cleaved-caspase-3. |
