目的 建立高效液相色谱法波长切换法同时测定陈皮配方颗粒中橙皮苷、川陈皮素和橘皮素的含量。方法 采用Diamonsil C18色谱柱(250 mm×4.6 mm,5 μm);流动相为乙腈(A)-0.1%醋酸水(B),二元梯度洗脱(0~6 min,15%A→20%A;6~16 min,20%A→34%A;16~20 min,34% A→52%A;20~28 min,52% A;28~30 min,52%A→100%A;30~32 min,100%A→15%A),流速1.0 mL·min-1,柱温30 ℃,波长切换(0~16 min,在283 nm波长下检测橙皮苷;16~ 32 min,在330 nm波长下检测川陈皮素和橘皮素)。结果 橙皮苷、川陈皮素和橘皮素的进样量分别在120.180~2 403.600 ng(r=0.999 8)、9.883~197.660 ng (r=0.999 6)和4.720~94.400 ng (r=0.999 5)范围内呈良好的线性关系;平均回收率(n=6)分别为98.29%、98.43%、97.74%,RSD分别为2.48%、1.34%、2.05%。结论 经方法学验证,该法可用于陈皮配方颗粒中3种黄酮类成分的含量测定。 |

Objective To develop an HPLC method for simultaneous determination of hesperidin,nobiletin and tangeretin in Pericarpium Citri Reticulatae Dispensing Granules.Methods The HPLC system consisted of a Diamonsil C18column(250 mm×4.6 mm,5 μm) and the mobile phase of acetonitrile(A)-0.1%(v/v)acetic acid water(B)in gradient mode(0-6 min,15%A→20%A;6-16 min,20%A→34%A;16-20 min,34% A→52%A;20-28 min,52 % A;28-30 min,52%A→100%A;30-32 min,100%A→15%A)with a flow rate of 1.0 mL·min-1 at 30 ℃.The detection wavelength was 283 nm(0-16 min)for hesperidin,330nm(16-32 min)for nobiletin and tangeretin.Results The contents of hesperidin,nobiletin and tangeretin had good linear relationship in the ranges of 120.180-2 403.600 ng(r=0.999 8),9.883-197.660 ng(r=0.999 6),and 4.720-94.400 ng(r= 0.999 5)respectively,and the average recoveries(n=6)of five components were 98.29 % with RSD of 2.48 %,98.43 % with RSD of 1.34 % and 97.74 % with RSD of 2.05% respectively.Conclusion According to the methodology validation,this method can be applied to the determination of hesperidin,nobiletin and tangeretin in Pericarpium Citri Reticulatae Dispensing Granules simultaneously. |
