      目的 研究桂枝茯苓胶囊对大鼠子宫肌瘤中孕激素受体(PR)和子宫平滑肌细胞蛋白激酶C(PKC)的影响。方法 正常组:14只,肌注0.9%生理盐水0.05 mL(3次/周);造模组:67只,Wistar雌性大鼠注射黄体酮、苯甲酸雌二醇建立子宫肌瘤模型,分别予以桂枝茯苓流浸膏和米非司酮灌胃,用免疫组化法测定药物对各组大鼠子宫平滑肌组织中PR及PKC。结果 ①用药后表明,PR阳性表达率,桂枝茯苓组较模型组下降显著[(32.25±2.8)%比(75.25±4.1)%,P<0.05],接近正常组水平[(32.25±2.8)%比(28.34±2.4)%,P>0.05]。②PKC细胞膜的活性表达,桂枝茯苓组较模型组下降显著[(21.62±5.23)比(37.24±7.12) fmol·mg-1·min-1,P<0.05],桂枝茯苓组更接近正常组[(21.62±5.23)比(19.31±3.84) fmol·mg-1·min-1,P>0.05]。③PKC细胞质的活性表达,桂枝茯苓组与模型组比较升高明显[(16.50±2.80)比(11.68±4.68) fmol·mg-1·min-1,P<0.05],桂枝茯苓组PKC细胞质的活性升高接近正常组表达[(16.50±2.80)比(15.43±4.59) fmol·mg-1·min-1,P>0.05]。结论 桂枝茯苓胶囊显著改善子宫平滑肌细胞的缺氧状态,抑制血管生成,切断子宫平滑肌细胞增殖分化信号传递通道,促使子宫平滑肌细胞的修复,这可能是桂枝茯苓胶囊抑制子宫肌瘤发展的重要机制之一。

      Objective To observe the effect of Guizhi Fuling capsule on progesterone receptor(PR) and vascular smooth muscle cell protein kinase C (PKC) in rat uterine leiomyoma.Methods Wistar female rats were injected with Estradiol benzoate and Progesterone to establish an animal model of uterine leiomyoma.Both?group? were?given?Guizhi Fuling fluidextractum and Mifepristone by?gavage.The effects of Guizhi Fuling to PR and vascular smooth muscle cells PKC in smooth muscle tissue of rats were determined by immunehistochemical.Results ① After administration,compared with the model group,positive expression rate of PR were decreased significantly,the Guizhi Fuling group [(32.25±2.8)% vs.(75.25±4.1)%,P<0.05],close to the normal group [(32.25±2.8)% vs.(28.34±2.4)%,P>0.05].② Active expression of PKC cell membrane,compared with the model group,the Guizhi Fuling group decreased significantly[(21.62±5.23) vs.(37.24±7.12) fmol·mg-1·min-1,P<0.05],the Guizhi Fuling group was closer to the normal group[(21.62±5.23) vs.(19.31±3.84) fmol·mg-1·min-1,P>0.05].③ The active expression of PKC cytoplasm,compared with the model group,the Guizhi Fuling group increased significantly [(16.50±2.80) vs.(11.68±4.68) fmol·mg-1·min-1,P<0.05],the activity of cytoplasm of PKC in Guizhi Fuling group was higher than that in normal group [(16.50±2.80) vs.(15.43±4.59) fmol·mg-1·min-1,P>0.05].Conclusion Guizhi Fuling capsule can significantly improve the hypoxia status of uterine smooth muscle cells,inhibite angiogenesis,cut off the signal transduction pathway of proliferation and differentiation of uterine smooth muscle cells,promote the repair of uterine smooth muscle cells,which may be one of the important mechanisms of inhibiting the development of uterine fibroids by Guizhi Fuling capsule.
