      目的 探讨血清25羟维生素D水平与年龄相关性白内障的关系。方法 将Wistar大鼠采用随机数字表法分为正常对照组、年龄相关性白内障模型组、维生素D低剂量组和维生素D高剂量组。采用D-半乳糖(50%浓度)诱导构建年龄相关性白内障大鼠模型,应用眼压计及裂隙灯显微镜检测大鼠眼压及晶状体浑浊情况;流式细胞仪检测晶状体上皮细胞凋亡情况;通过聚合酶链式反应(RT-PCR)法检测大鼠晶状体上皮细胞中DPPⅡ、c-mycmRNA表达;应用免疫印迹(Western Blot)检测肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)蛋白表达量。结果 与空白对照组相比,白内障模型组25羟维生素D水平降低,眼压升高;维生素D给药后,维生素D水平较模型组升高,眼压降低(25羟维生素D水平:F=9.215,P=0.007;眼压:F=12.502,P=0.005)。与空白对照组相比,白内障模型组25羟维生素D水平降低,眼压升高;维生素D给药后,维生素D水平较模型组升高,眼压降低(25羟维生素D水平:F=9.215,P=0.007;眼压:F=12.502,P=0.005)。各组LEC细胞凋亡率均随时间延长而缓慢增加;白内障模型组晶体上皮细胞凋亡率高于空白对照组(F=8.212,P=0.016)。给药后,血清中维生素D水平有一定程度的恢复,晶状体浑浊程度降低(F=11.635,P=0.003)。给药后,血清中维生素D水平有一定程度的回升,LED凋亡程度降低(1周:F=10.258,P=0.0111;2周:F=13.390,P=0.005;4周:F=14.157,P=0.003)。RT-PCR结果显示,维生素D对照组大鼠晶状体上皮细胞织中的DPPⅡ、c-myc低于模型组(DPPⅡ:F=11.217,P=0.008;c-myc:F=13.087,P=0.005);Western Blot检测结果表明,血清中中25羟维生素D可下调TNF-α、IL-8蛋白表达水平(DPPⅡ:F=12.023,P=0.002;c-myc:F=10.419,P=0.005)。结论 血清中25羟维生素D水平与年龄相关性白内障的发展负相关,这对于白内障的预防及治疗有十分重要的意义。

      Objective The relationship between serum 25-hydroxyvitamin D level and age-related cataract was studied.Methods Wistar rats were randomly divided into blank control group,model group and vitamin D administration group.D-galactose (50% concentration) was used to induce the construction of age-associated cataract rat model.The apoptosis of lens epithelial cells was detected by flow cytometry.The rat lens epithelial cells in the DPP Ⅱ,c-mycmRNAexpression weredetect by RT-PCR.TNF-α and il-8 protein expression were detected by Western Blot.Results Compared with the blank control group,the level of 25-hydroxyvitamin D in cataract model group decreased and the intraocular pressure increased.After vitamin D administration,the level of vitamin D increased and the intraocular pressure decreased (25-hydroxyvitamin D level:F=9.215,P=0.007;intraocular pressure:F=12.502,P=0.005).Compared with the blank control group,the level of 25-hydroxyvitamin D in cataract model group decreased and the intraocular pressure increased.After vitamin D administration,the level of vitamin D increased and the intraocular pressure decreased (25-hydroxyvitamin D level:F= 9.215,P=0.007;intraocular pressure:F=12.502,P=0.005).The apoptotic rate of LEC cells in each group increased slowly with time.The apoptotic rate of lens epithelial cells in cataract model group was higher than that in blank control group (F=8.212,P=0.016).After administration,the level of vitamin D in serum recovered to a certain extent,and the degree of lens opacity decreased (F=11.635,P=0.003).After administration,the level of vitamin D in serum increased to some extent and the apoptotic degree of LED decreased (1 week:F=10.258,P=0.0111;2 weeks:F=13.390,P=0.005;4 weeks:F=14.157,P=0.003).RT-PCR results showed that DPP Ⅱ and c-myc in vitamin D control group were lower than those in model group (DPP Ⅱ:F=11.217,P=0.008;c-myc:F=13.087,P=0.005).Western Blot results showed that serum 25-hydroxyvitamin D could down-regulate the expression of TNF-a and IL-8 (DPP Ⅱ:F=12.023,P=0.002;c-myc:F=10.419,P=0.005).Conclusion The serum level of 25-hydroxyvitamin D is negatively correlated with the development of age-related cataract,which is of great significance for the prevention and treatment of cataract.
