      目的探讨分析肝细胞生长因子( HGF)/酪氨酸蛋白激酶 MET(c-met)信号通路与乳腺癌侵袭转移的相关性。方法选择 2018年 3月至 2019年 3月承德医学院附属医院临床术后经病理证实的乳腺癌标本 75例、乳腺纤维腺瘤标本 60例以及癌旁正常乳腺组织标本 50例。采用 SP免疫组化法以及逆转录聚合酶链反应( RT-PCR)法检测各组织标本 HGF、c-met、β连环素(β-catenin)蛋白表达和 mRNA表达情况。结果 HGF[82.67%(62/75)比 48.33%(29/60)比 14.00%(7/50)]、 c-met[86.67%(65/ 75)比 55.00%(33/60)比 20.00%(10/50)]、 β-catenin[78.67%(59/75)比 41.67%(25/60)比 6.00%(3/50)]在乳腺癌组织、乳腺纤维腺瘤组织以及癌旁正常乳腺组织中的表达均差异有统计学意义(P<0.05)。在乳腺癌、乳腺纤维腺瘤以及癌旁正常乳腺组织中 HGF、c-met mRNA相对表达量差异有统计学意义( P<0.05),其中乳腺癌相对表达量最高、乳腺纤维腺瘤相对表达量次之;而乳腺癌、乳腺纤维腺瘤以及癌旁正常乳腺组织中 β-catenin mRNA相对表达量比较差异无统计学意义(P>0.05)。HGF表达阳性率与病人 TNM分期、淋巴结转移、肿瘤大小、 ER表达、 HER-2表达有关( P<0.05)。 c-met蛋白表达阳性率与病人肿瘤 TNM分期、淋巴结转移密切相关( P<0.05)。 β-catenin蛋白表达异常率与病人 TNM分期、淋巴结转移、肿瘤大小密切相关( P<0.05)。结论乳腺癌组织中 HGF、c-met蛋白高表达而 β-catenin蛋白异常表达,且 HGF/c-met信号通路及 β-catenin蛋白与乳腺癌侵袭转移密切相关。

      Objective To investigate the correlation between HGF/c-met signaling pathway and invasion and metastasis of breast cancer.Methods From March 2018 to March 2019, 75 breast cancer specimens, 60 breast fibroadenoma specimens and 50 normalbreast tissue specimens were selected from the Affiliated Hospital of Chengde Medical College. The expression of HGF, c-met and βcatenin protein and the expression of HGF, c-met and β-catenin mRNA were detected by SP immunohistochemistry and RT-PCR.Re? sults There were significant differences in the expression of HGF[82.67%(62/75) vs. 48.33%(29/60) vs. 14.00%(7/50)], c-met[86.67% (65/75) vs. 55.00%(33/60) vs. 20.00%(10/50)] and β-catenin[78.67%(59/75) vs. 41.67%(25/60) vs. 6.00%(3/50)] in breast cancer, breast fibroadenoma and adjacent normal breast tissues (P<0.05). The relative expression of HGF and c-Met mRNA in breast cancer, breast fi? broadenoma and adjacent normal breast tissues were significantly different (P<0.05), and the relative expression of HGF and c-Met mRNA in breast cancer was the highest, followed by that in breast fibroadenoma. There was no significant difference in the relative ex?pression of β-catenin protein between breast cancer, breast fibroadenoma and adjacent normal breast tissues (P>0.05). The positive rate of HGF expression was related to TNM stage, lymph node metastasis, tumor size, ER expression and HER-2 expression (P<0.05). The positive rate of c-met protein expression was closely related to TNM stage and lymph node metastasis (P<0.05). The abnormal ex? pression rate of β– catenin protein was closely related to TNM stage, lymph node metastasis and tumor size (P<0.05).Conclusions The expression of HGF and c-Met protein in breast cancer tissues is up-regulated while that of β-catenin protein is abnormal. HGF/cmet signaling pathway and β-catenin protein are closely related to the invasion and metastasis of breast cancer.
