目的探究紫花前胡苷抗宫颈癌 HeLa细胞的作用效果及相关信号通路。方法 HeLa细胞在 0、0.1、1和 10 mmol/L浓度的紫花前胡苷作用 24 h后,通过 CCK-8法检测细胞增殖活性,流式细胞术检测细胞凋亡水平;通过蛋白免疫印迹法检测细胞内凋亡相关 cleaved-caspase 3(cle-caspase 3)、Cytochrome C(Cyto C)、Bax、Bcl2蛋白水平变化,以及自噬相关 p-mTOR、mTOR、 P62、Beclin1、LC3蛋白水平变化;结合 Ad-GFP-LC3B腺病毒转染检测细胞内 LC3B聚集状态。结果对照组(0 mmol/L)、0.1 mmol/L组、1 mmol/L组、10 mmol/L组的 HeLa细胞凋亡率依次升高,分别为(14.93±2.95)%、(15.43±2.13)%、(21.63±3.19)%、 |

Objective To investigate the effect and related signaling of Nodakenin against cervical cancer HeLa cells.Methods HeLa cells were treated with Nodakenin under concentration of 0, 0.1, 1 and 10 mmol/L for 24 h. Cell viability was detected by CCK-8 assay, and cell apoptosis was detected by Flow cytometry. Protein levels of apoptosis related cle-caspase 3, Cyto C, Bax and Bcl2, as well as protein levels of autophagy related p-mTOR, mTOR, P62, Beclin1 and LC3 were detected by western blot.Intracellular LC3B aggregation was detected by adenovirus transfection with Ad-GFP-LC3B.Results The viability of HeLa cells was decreased and the apoptosis was increased by Nodakenin in a dose-dependent manner, and 10 mmol/L is the most significant concentration(P<0.05). After the treatment of 10 mmol/L Nodakenin for 24 hours, the protein levels of pro-apoptotic proteins cleaved-caspase 3(cle-caspase 3), Cytochrome C(Cyto C) and Bax increased, while the protein levels of anti-apoptotic protein Bcl2 decreased, with statistically significant differences(P<0.05). Besides, the ratio of autophagy related p-mTOR/mTOR decreased, the accumulation of Beclin1 and LC3-Ⅱ increased, and levels of autophagy flow-related protein P62decreased. In addition, LC3B presented granular aggregation in the cells of Nodakenin-treated group(P<0.05).Conclusion Nodakenin exhibits excellent anti-tumor effects in cervical cancer HeLa cells, and the underlying mechanism maybe related to autophagy related cell apoptosis. |
