| 目的探讨 siRNA沉默人多梳基因 2(HPC2)对 5-8F鼻咽癌细胞增殖的作用和细胞周期的影响。方法用 RNAi MAX将靶向 HPC2的 siRNA转染入鼻咽癌细胞 5-8F，实验分 scrambled阴性对照组， #1 siRNA-HPC2组， #2 siRNA-HPC2组，用 CCK8法比较细胞增殖活力，平板克隆实验比较增殖能力，流式细胞仪检测比较各组细胞周期分布。结果 siRNA-HPC2细胞转染组的 mRNA及蛋白表达较对照组明显降低( P <0.05)，两个 HPC2沉默组的细胞增殖活性及克隆形成能力较对照组明显下降( P< 0.05)。流式细胞检测发现 HPC2沉默组的细胞 G1/S时相转换较对照组减缓。结论沉默 HPC2表达，可以减弱鼻咽癌细胞的恶性增殖，与干扰了细胞周期的运行有关。 |

| Objective To investigate the effect of HPC2 silenced by siRNA on the proliferation and the influence of cell cycle of 58F NPC cells.Methods RNAi Max was used to transfect siRNA targeting HPC2 into NPC cell line 5-8F. The experiment was divided into three groups: negative control group, #1 siRNA-HPC2 group and #2 siRNA-HPC2 group.Results Expression of mRNA and protein in the siRNA-HPC2 cell transfection groups were significantly lower than that in the control group (P < 0.05). Cell proliferation viability and clone formation ability in the two HPC2 silenced groups were obviously decreased than that in the control group (P < 0.05).Flow cytometry detection demonstrated that G1 / S phase transition in HPC2 silenced groups were slower than that in control group.Conclusion Silencing the expression of HPC2 by siRNA can inhibit the malignant proliferation of NPC cells, which is related to thecell cycle impaired. |
