目的探讨甲状腺组织和外周血单个核细胞中微小 RNA-142-3p(miR-142-3p)、微小 RNA-150(miR-150)在桥本甲状腺炎( HT)中的表达水平及临床意义。方法选取 2017年 3月至 2019年 6月南阳市中心医院行甲状腺手术切除并经病理确诊为 HT病人 87例为 HT组;同期选取南阳市中心医院行甲状腺腺瘤切除术的非癌病人 73例为对照组。采用实时荧光定量 PCR(qRT-PCR)法检测两组甲状腺组织和外周血单个核细胞中 miR-142-3p、miR-150表达水平;分析 HF病人 miR-142-3p、miR-150水平与甲状腺功能参数及相关细胞因子相关性。结果 HT组甲状腺组织和外周血单个核细胞中 miR-142-3p[( 1.54±0.32)、(2.25±0.54)]比[( 1.00±0.23)、(1.00±0.24)]、 miR-150[( 2.21±0.43)、(3.78±1.14)]比[( 1.00±0.16)、(1.00±0.19)]表达水平均显著高于对照组( P<0.05);两组甲状腺功能参数及相关细胞因子水平比较,均差异有统计学意义, HT组促甲状腺激素( TSH)、状腺过氧化物酶抗体( TPOAb)、白细胞介素 17(IL-17)、白细胞介素 6(IL-6)、干扰素 -γ(IFN-γ)、转化生长因子 β1(TGF-β1)水平甲显著高于对照组( P<0.05),游离三碘甲状腺原氨酸(FT3)、游离甲状腺素( FT4)水平显著低于对照组( P<0.05); HT病人甲状腺组织和外周血单个核细胞中 miR-142-3p、miR-150水平呈正相关,且与 TSH、TPOAb、IL-17、IL-6、IFN-γ、TGF-β1呈正相关,与 FT3、 FT4呈负相关( P<0.05)。结论甲状腺组织和外周血单个核细胞中 miR-142-3p、miR-150水平的变化与 HT有关。 miR-142-3p、 miR-150对评价 HT的早期危险性、降低患病率有重要意义。 |

Objective To detect the expression levels of microRNA-142-3p (miR-142-3p) and microRNA-150 (miR-150) in thyroidtissues and peripheral blood mononuclear cells of Hashimoto's thyroiditis (HT) patients, and to explore the clinical significance of theirexpression levels in HT patients.Methods A total of 87 patients, who were hospitalized in Nanyang Central Hospital for thyroidectomy and diagnosed as HT by pathology from March 2017 to June 2019, were selected as the study objects (HT group). And 73 cases ofnon-cancer patients, who underwent thyroidectomy in Nanyang Central Hospital during the same time, were selected as the controlgroup (control group). The expression levels of miR-142-3p and miR-150 in thyroid tissues and peripheral blood mononuclear cells were detected by real-time fluorescence quantitative PCR (qRT-PCR). The correlations between the levels of miR-142-3p, miR-150 and thyroid function parameters, related cytokines in HF patients were analyzed.Results The expression levels of miR-142-3p [(1.54± 0.32), (2.25±0.54)] vs. [(1.00±0.23), (1.00±0.24)] and miR-150 [(2.21±0.43), (3.78±1.14)] vs. [(1.00±0.16), (1.00±0.19)] in thyroid tissue and peripheral blood mononuclear cells in HT group were significantly higher than those in control group (P < 0.05). There weresignificant differences in thyroid function parameters and related cytokines between the two groups. The levels of thyroid stimulatinghormone (TSH), thyroid peroxidase antibody (TPOAb), interleukin-17 (IL-17), interleukin-6 (IL-6), interferon-γ (IFN-γ) and transforming growth factor β1 (TGF-β1) in HT group were significantly higher than those in control group (P<0.05). The levels of free triiodothyronine (FT3) and free thyroxine (FT4) were significantly lower in HT group than those in control group (P<0.05). The levels of miR-1423p and miR-150 in thyroid tissue and peripheral blood mononuclear cells of HT patients were positively correlated, and they were alsopositively correlated with TSH, TPOAb, IL-17, IL-6, IFN-γ, TGF-β 1, while negatively correlated with FT3 and FT4 (P < 0.05).Conclu? sions The changes in the levels of miR-142-3p and miR-150 in thyroid tissue and peripheral blood mononuclear cells are related to HT. MiR-142-3p and miR-150 have great significance in evaluating the early risk of HT and reducing the prevalence. |
