      目的研究长链非编码 RNA PCGEM1(lncRNA PCGEM1)与信号传导转录激活因子 3(STAT3)在卵巢癌中的表达及意义。方法选取 2013年 3月至 2014年 12月南阳市第一人民医院行卵巢摘除手术的卵巢癌病人 108例,术中留取卵巢组织,选取行手术切除的卵巢囊肿病人 108例,术中留取已证实无肿瘤细胞的正常卵巢组织,采用实时荧光定量 PCR(RT-qPCR)检另测卵巢癌及正常卵巢组织中 lncRNA PCGEM1、STAT3 mRNA表达水平,采用免疫组化法检测 STAT3蛋白阳性表达率,采用 Pearson法进行相关性分析,采用 Kaplan-Meier生存曲线分析 lncRNA PCGEM1、STAT3蛋白表达与病人 5年生存率的关系;并以 Cox回归分析影响病人预后的危险因素。结果与正常卵巢组织比较,卵巢癌组织 lncRNA PCGEM1[( 2.41±0.43)比( 0.99±0.15)]、 STAT3 mRNA表达水平[(2.69±0.52)比(1.02±0.16)]、 STAT3蛋白阳性表达率( 70.37%比 35.19%)显著升高(P<0.05);卵巢癌组织 lncRNA PCGEM1、STAT3蛋白阳性表达与病人年龄无关( P>0.05)与 FIGO分期、病理分级、铂类耐药及淋巴结转移有关( P<0.05)。卵巢癌组织 lncRNA PCGEM1与 STAT3 mRNA表达水平呈关( P<0.05)。 lncRNA PCGEM1高表达组 5年生存率为 8.33%,显著低于低表达组 54.35%(χ2=39.51,P<0.001),STAT3蛋白阳性表达组 5年生存率为 13.16%,显著低于阴性表达组 68.75%(χ2=26.16,P<0.001)。顺铂耐药、淋巴结转移、 lncRNA PCGEM1高表达、 STAT3蛋白阳性表达是影响卵巢癌病人预后的独立危险因素(均 P<0.05)。结论卵巢癌组织中 lncRNA PCGEM1、STAT3均高表达,且二者呈正相关,与卵巢癌病人病情及 5年生存率有关,有望成为评估病人预后的生物标志。

      Objective To study the expressions and significances of long non-coding RNA PCGEM1 (lncRNA PCGEM1) and signal transduction activator 3 (STAT3) in ovarian cancer.Methods A total of 108 patients with ovarian cancer who underwent ovariectomyin the First People's Hospital of Nanyang from March 2013 to December 2014 were selected, ovarian tissue was retained during the operation, and 108 patients with ovarian cysts who underwent surgical resection were selected, and normal ovarian tissues without tumorcells were retained during the operation. The expression levels of lncRNA PCGEM1 and STAT3 in ovarian cancer and normal ovariantissues were detected by real-time quantitative PCR (RT-qPCR).The positive expression rate of STAT3 protein was detected by immunohistochemistry. Pearson method was used for correlation analysis, Kaplan-Meier survival curve was used to analyze the relationships between lncRNA PCGEM1, STAT3 protein expression and the 5-year survival rate of patients, and Cox regression analysis was used to analyze the risk factors affecting the prognosis of patients.Results Compared with normal ovarian tissues, the mRNA levels of lncRNA PCGEM1[(2.41±0.43) vs. (0.99±0.15)], STAT3 [(2.69±0.52) vs.(1.02±0.16)] and the positive rate (70.37% vs.35.19%) of STAT3 protein in ovarian cancer tissues were significantly increased (P<0.05). The positive expressions of lncRNA PCGEM1 and STAT3 proteins in ovarian cancer tissues were not related to age (P>0.05), but related to FIGO stage, pathological grade, platinum resistance and lymph node metastasis (P<0.05). The expression levels of lncRNA PCGEM1 and STAT3 in ovarian cancer tissues were positively correlated (P <0.05). The 5-year survival rate of the high expression group of lncRNA PCGEM1 was 8.33%, which was significantly lower than54.35% of the low expression group (χ2= 39.51,P<0.001), and the 5-year survival rate of the STAT3 positive expression group was13.16%, which was significantly lower than 68.75% of the negative expression group (χ2=26.16, P<0.001). Cisplatin resistance, lymphnode metastasis, high expression of lncRNA PCGEM1 and positive expression of STAT3 protein were independent risk factors affectingthe prognosis of ovarian cancer patients (all P<0.05).Conclusion The lncRNA PCGEM1 and STAT3 are highly expressed in ovariancancer tissues, and they are positively correlated with the condition and 5-year survival rate of ovarian cancer patients, which are expected to become biomarkers to evaluate the prognosis of patients.
