      目的通过基因共表达网络分析( weighted gene co-expression network analaysis,WGCNA)及蛋白质相互作用( protein-protein interaction,PPI)识别肺腺癌发生发展中的枢纽基因。方法从 TCGA数据库中下载 497例肺腺癌组织和 54例正常肺组织的 RNA-seq表达矩阵及配套临床信息。利用 R语言的 DESeq2软件包进行差异基因表达分析。随后通过 WGCNA筛选出枢纽模块, PPI分析鉴定出枢纽基因。最后使用 GEIPA数据库验证枢纽基因与肿瘤分期、预后的关系。结果通过 DESeq2分析得到 1 904个差异基因。通过构建共表达网络,确定棕色和蓝色模块为枢纽模块, PPI分析筛选得到核分裂周期蛋白 80(nu. clear division cycle 80,NDC80)为枢纽基因。 GEIPA数据库验证结果显示 NDC80基因与肿瘤的分期相关( F=3.58,P<0.05),且 NDC80基因高表达与肿瘤的总生存期较差有明显相关( HR=1.6,P<0.05)。结论通过构建 WGCNA及 PPI分析得到枢纽基因 NDC80,且 NDC80基因的高表达与肿瘤的分期及预后密切相关。提示 NDC80基因有望成为肺腺癌预后的生物学标志物。
英文摘要:
      Objective To identify the hub gene associated with development and progression of lung adenocarcinoma using weight. ed gene co-expression network analysis (WGCNA) and protein-protein interaction (PPI) network analysis.Methods The RNA-seq ex. pression matrix data and patient-related clinical data from 497 lung adenocarcinoma tissues and 54 normal lung tissues were download.ed from the TCGA database. The difference of gene expression was analyzed by DESeq2 software package in R language. Then, the hubmodules were identified using WGCNA, and the hub gene was identified using PPI network. Finally, GEIPA database was used to veri.fy the correlations between the expression levels of the hub gene and tumor stage and prognosis.Results A total of 1904 differential genes were obtained by the DESeq2 analysis. Through constructing the co-expression network, the brown module and blue module wereidentified as the hub modules, then, the hub gene nuclear division cycle 80 (NDC80) was screened out using PPI network. The valida.tion results of GEIPA database showed that NDC80 was associated with tumor stage (F=3.58, P<0.05), and increased NDC80 was sig. nificantly related to poor overall survival (HR=1.6,P<0.05).Conclusion The hub gene NDC80 was identified by WGCNA and PPIanalysis, which were associated with tumor stage and prognosis.This suggests that the NDC80 may be a biomarker for the prognosis oflung adenocarcinoma.
