      目的探究茯苓多糖( PAC)对 2型糖尿病( T2DM)大鼠丝裂原激活的蛋白激酶( MAPK)通路及胰岛素抵抗( IR)的影响。方法 2019年 3—10月,采用高脂高糖喂养联合腹腔注射链脲佐菌素( STZ)建立 T2DM大鼠模型,采用随机数字表法分为模型( T2DM)组、二甲双胍( MET)组( 0.65 g·kg.1·d.1)和 PAC低( PAC-L,0.05 g·kg.1·d.1)、中( PAC-M,0.10 g·kg.1·d.1)、高剂量(PAC-H,0.20 g·kg.1 ·d.1)组,每组 12只。另取 12只正常非 T2DM大鼠为正常组( NC组)干预 8周后测定空腹血糖、空腹血清胰岛素( FINS)、三酰甘油及肝组织均浆丙二醛、谷胱甘肽过氧化物酶( GSH-Px)和超氧化物,歧化酶( SOD)活性,采用蛋白质印迹法检测肝脏组织磷酸化 p38丝裂原激活的蛋白激酶( p-p38MAPK)、磷酸化 c-Jun氨基末端激酶( p-JNK)蛋白表达水平。结果与 NC组比较, T2DM组大鼠血清空腹血糖、 FINS、三酰甘油含量、胰岛素抵抗指数( HOMA-IRI)(62.56±5.11比 5.81±1.05)、肝脏组织丙二醛含量、 p-p38MAPK(1.35±0.08比 0.16±0.03)和 p-JNK(1.29±0.08比 0.17±0.02)蛋白水平均显著升高( P<0.05)胰岛素敏感指数( ISI)[( 0.86±0.05)×10.3比( 7.69±0.14)×10.3]、肝脏组织 GSH-Px、SOD活性均显著降低( P<0.05);与 T2DM组比,较, PAC-L组、 PAC-M组、 PAC-H组大鼠血清空腹血糖、 FINS、三酰甘油含量、 HOMA-IRI(49.47±4.56、31.14±3.03、26.59±2.16比

      Objective To investigate the effect of pachyman (PAC) on mitogen activated protein kinase (MAPK) pathway and insulinresistance (IR) in type 2 diabetic rats.Methods From March 2019 to October 2019, the T2DM rat model was established by high fatand high sugar feeding combined with intraperitoneal injection of streptozotocin (STZ). The rats were randomly divided into model(T2DM) group, metformin (MET) group (0.65g·kg.1·d.1) and PAC low (PAC-L, 0.05 g·kg.1·d.1), medium(PAC-M, 0.10 g·kg.1·d.1) and high (PAC-H, 0.20 g·kg.1·d.1) dose group, with 12 rats in each group. Another 12 non-T2DM rats were taken as normal group (NC group), After 8 weeks of intervention, the levels of fasting blood-glucose, fasting serum insulin (FINS) , triglyceride, the activities of glu.tathione peroxidase (GSH PX), superoxide dismutase (SOD) and malondialdehyde were determined, the protein expression levels ofphosphorylated p38 mitogen activated protein kinase (p-p38MAPK) and phosphorylated c-Jun amino terminal kinase (p-JNK) in liver tissue were detected by Western blot.Results Compared with NC group, the levels of serum fasting blood-glucose, FINS, triglyceride, HOMA-IRI (62.56±5.11 vs. 5.81±1.05), malondialdehyde content of liver tissue, p-p38 MAPK (1.35±0.08 vs. 0.16±0.03) and p-JNK (1.29±0.08 vs. 0.17±0.02) protein in T2DM group increased significantly (P<0.05), while ISI [(0.86±0.05)×10.3 vs. (7.69±0.14)×10.3], GSH-Px of liver tissue and SOD activity decreased significantly (P<0.05); compared with T2DM group, the levels of serum FBG, FINS, triglyceride, HOMA-IRI (49.47±4.56, 31.14±3.03, 26.59±2.16 vs. 62.56±5.11), malondialdehyde content of liver tissue, p-p38 MAPK (1.04±0.06, 0.89±0.03, 0.39±0.02 vs. 1.35±0.08) and p-JNK (0.85±0.02, 0.72±0.05, 0.31±0.01 vs. 1.29±0.08) in PAC-L group, PAC-M group and PAC-H group decreased significantly (P<0.05), while ISI [(1.51±0.11)×10.3, (2.14±0.12)×10.3, (3.73±0.15)×10.3 vs. (0.86± 0.05)×10.3], GSH-Px of liver tissue and SOD activity increased significantly (P < 0.05), which was dose-dependent (P<0.05). PAC-H group was better than MET group (P < 0.05).Conclusion PAC can improve insulin resistance in T2DM rats, which may be related to its effects in inhibiting MAPK pathway and reducing oxidative stress.
