      目的探讨芦丁对右旋葡聚糖硫酸钠( DSS)诱导的大鼠溃疡性结肠炎( UC)的缓解作用以及对核因子 κB(NF-κB)信号通路的影响。方法 2020年 4—5月建立右旋葡聚糖硫酸钠( DSS)诱导的 UC模型。将大鼠分为对照组、模型组和芦丁组( 20、 40、80 mg/kg)。观察各组大鼠的疾病活动指数( DAI),比色法检测结肠组织髓过氧化物酶( MPO)活性, Elisa法检测大鼠结肠组织中的肿瘤坏死因子 -α(TNF-α)和白细胞介素 -6(IL-6)水平,并采用蛋白质印迹法( western blotting)测定各组大鼠结肠组织的人磷酸化 IκB激酶 -α(p-IκBα)、 IκB激酶 -α(IκBα)以及核 p65蛋白表达。结果芦丁高、中、低剂量组的 DAI评分[(1.09±0.39)、(1.38±0.38)、(1.97±0.46)分]、 MPO活性[( 0.74±0.04)、(0.75±0.08)、(0.98±0.06)U/g]、 TNF-α水平[( 3.16±0.52)、(3.75±0.72)、(4.26±0.87)μg/L]和 IL-6水平[( 3.72±0.65)、(4.28±0.72)、(5.12±0.93)μg/L]均显著性低于模型组的( 2.98±0.57)分、(1.26±0.07)U/g、(5.19±0.93)μg/L、(6.54±0.85)μg/L(P<0.05)。机制研究显示芦丁可以有效下调 DSS诱导大鼠结肠组织的 p-IκBα和核 p65的水平的提高(P<0.05)。结论芦丁通过下调 NF-κB信号通路缓解右旋葡聚糖硫酸钠诱导的大鼠溃疡性结肠炎。

      Objective To investigate the relief effect of rutin on dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) in rats and its effect on nuclear factor kappa-B (NF-κB) signaling pathway.Methods A rat model of Dextran Sulfate Sodium Salt (DSS)-induced UC was established from April 2020 to May 2020. Rats were assigned into control group, model group and rutin group (20 mg/kg,40 mg/kg, 80 mg/kg). The disease activity index (DAI) of rats in each group was observed, myeloperoxidase (MPO) activity of colon tissue was detected using colorimetric method, tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in rat colon tissue were detected using Elisa method, and p-IκBα, IκBα and nuclear p65 protein in colon tissue were determined using Western blot.Results The DAI score [(1.09±0.39), (1.38±0.38), (1.97±0.46)], MPO activity [(0.74±0.04), (0.75±0.08), (0.98±0.06)U/g], TNF-α levels [(3.16±0.52), (3.75±0.72), (4.26±0.87) μg/L] and IL-6 levels [(3.72±0.65), (4.28±0.72), (5.12±0.93) μg/L] of high, middle, low rutin group were allsignificantly lower than those of model group [(2.98±0.57), (1.26±0.07)U/g, (5.19±0.93) μg/L, (6.54±0.85) μg/L] (P<0.05). The mechanism study showed that rutin can effectively down-regulate DSS induced increase of p-IκBα and nuclear p65 in rat colon tissue (P< 0.05).Conclusion Rutin alleviates dextran sodium sulfate-induced ulcerative colitis in rats by down-regulating the NF-κB signaling pathway.
