      目的探讨千金藤素对肝癌细胞 HepG2增殖、凋亡的影响和机制。方法 2020年 1—6月,从上海通派生物科技有限公司购入肝癌细胞 HepG2,用千金藤素( 20 μmol/L)处理肝癌细胞 HepG2,MTT方法测定增殖活性, PI单染法检测细胞周期, Annexin V-FITC/PI双染法检测细胞凋亡,蛋白质印迹法( Western blotting)检测细胞周期蛋白 D1(cyclin D1)、 p21、Bcl-2相关 X蛋白( Bax)、活化胱天蛋白酶 3(cleaved caspase-3)、核因子 κB p65(NF-κB p65)蛋白表达。用 NF-κB信号激活剂和千金藤素联合处理肝癌细胞,检测细胞增殖、周期、凋亡变化。结果千金藤素处理后的肝癌细胞增殖活性下降,细胞 G0/G1期比例[(65.81±3.73)%比( 50.61±4.17)%]升高,细胞凋亡率[(13.47±1.58)%比( 2.18±0.12)%]升高,细胞中 p21、Bax、cleaved-caspase-3蛋白水平升高, cyclin D1蛋白水平下降, NF-κBp65蛋白水平( 0.19±0.02比 0.36±0.05)降低。 NF-κB信号激活剂处理可以提高千金藤素条件下肝癌细胞增殖活性( 0.46±0.03比 0.35±0.04)减少 G0/G1期比例,降低细胞凋亡率,降低细胞中 p21、Bax、cleavedcaspase-3蛋白表达水平,提高细胞中 cyclin D1和 NF-κBp65蛋,白水平。结论千金藤素抑制肝癌细胞增殖,阻滞细胞周期,诱

      Objective To investigate the effect and mechanism of cepharanthine on proliferation and apoptosis of hepatocellular carcinoma cell line HepG2.Methods From January 2020 to June 2020, HepG2 cells were purchased from Shanghai Tongpai Biotechnology Co., Ltd. HepG2 cells were treated with cepharanthine (20 μmol/L). MTT assay was used to determine the proliferation activity. Cellcycle was detected by PI monochrome staining. Annexin V-FITC/PI double staining was used to detect apoptosis. Western blotting was used to detect the expressions of cyclin D1, p21, Bcl-2-associated X protein (Bax), activated caspase 3 (cleaved caspase-3) and nuclear factor-κB p65 (NF-κBp65). Hepatocellular carcinoma cells were treated with NF-κB signal activator and cepharanthine to detect the changes of cell proliferation, cycle and apoptosis.Results The proliferative activity of hepatoma cells treated with cepharanthine decreased, the percentage of G0/G1 phase [(65.81±3.73) % vs. (50.61±4.17) %] increased, the apoptotic rate [(13.47±1.58) % vs. (2.18± 0.12) %] increased, the levels of p21, Bax and cleaved-caspase-3 protein in cells increased, Cyclin D1 protein level decreased, and the level of NF-κ Bp65 protein (0.19±0.02 vs. 0.36±0.05) decreased. NF-κB signal activator treatment could improve the proliferation (0.46±0.03 vs. 0.35±0.04) of hepatocellular carcinoma cells under the condition of cepharanthine, reduced the proportion of cells in G0/G1phase, decreased the apoptotic rate, decreased the expression levels of p21, Bax and cleaved-caspase-3 protein in cells, and increased the levels of cyclin D1 and NF-κBp65 in cells.Conclusion Cepharanthine inhibits the proliferation of hepatocellular carcinoma cells,blocks cell cycle and induces apoptosis, the mechanism of which is related to down-regulation of activation of NF-κB signaling.
