      目的探究新风胶囊调控巨噬细胞 M1/M2型极化对 DBA/1小鼠胶原诱导型关节炎( CIA)的影响。方法按体质量将 DBA/1小鼠采用随机数字表法分为空白对照组( NC组)、模型组( M组)和新风胶囊组( XFC组)三组。 M组和 XFC组建立 Ⅱ型胶原合并弗氏佐剂诱导的 CIA小鼠模型,第二次免疫当天, NC组和 M组给予生理盐水, XFC组按照规定剂量给予新风胶囊混悬液 1.5 mL/100 g,每天 1次,连续 21 d灌胃处理。利用小鼠关节炎指数( AI)评分评估新风胶囊对 CIA模型小鼠关节的作用, HE染色检测滑膜组织病理损伤程度;蛋白质印迹法( Western blotting)检测巨噬细胞中 CD68、iNOS蛋白表达水平。研究时间自 2020年 6月至 2021年 5月。结果实验结果表明,与 NC组相比,滑膜炎性细胞浸润数量及血管翳增生数量较前升高,第 30天 M组发病关节炎爪数[(3.83 ± 0.41)只]明显增多( P<0.05)右后爪关节炎严重程度评分[(2.83 ± 0.75)分]上升(P<0.05),四肢关节炎评分均值[( 2.17 ± 0.38)分]升高( P<0.05),CD68相对,表达量( 0.964±0.080)高于 NC组( 0.430±0.0186)(P<0.05)iNOS相对表达量(0.738±0.052)高于 NC组( 0.477±0.016)(P<0.05)。与 M组比较,第 30天 XFC组滑膜炎性细胞浸润数量及血管翳增,生数量均下降,发病关节炎爪数[(1.50 ± 0.55)只]低于 M组[(3.83 ± 0.41)只](P<0.05)右后爪关节炎严重程度评分[(1.00 ±

      Objective To investigate the effects of Xinfeng capsule on M1/M2 polarization of macrophages on collagen-induced ar-thritis (CIA) in DBA/1 mice.Methods According to body weight, DBA/1 mice were randomly divided into three groups: blank controlgroup (NC group), model group (M group) and Xinfeng capsule group (XFC group). CIA mouse model induced by typeⅡ collagen com-bined with fredrin's adjuvant was established in M group and XFC group. On the day of the second immunization, NC group and Mgroup were given normal saline, XFC group was given Xinfeng capsule suspension at the specified dose of 1.5 mL/100 g, once a day, for21 consecutive days. Arthritis Index (AI) score was used to evaluate the effects of Xinfeng capsule on CIA model mice joints, and HEstaining was used to detect the degree of synovial tissue pathological damage. The expression levels of CD68 and iNOS in macrophageswere detected by Western blotting. The study period was from June 2020 to May 2021.Results The results showed that compared withNC group, the number of synovial inflammatory cell infiltration and pannus hyperplasia increased, and the number of arthritic claws inM group [(3.83 ± 0.41)]significantly increased on day 30 (P<0.05), the severity score of right posterior paw arthritis [(2.83 ± 0.75) points] increased (P<0.05), the mean score of limb arthritis[(2.17 ± 0.38) points]increased (P<0.05), the relative expression of CD68 (0.964±0.080) was higher than that of NC group (0.430 ±0.0186)(P<0.05), the relative expression of iNOS in NC group (0.738 ± 0.052) was higher than that in NC group (0.477 ± 0.016) (P<0.05).Compared with M group, XFC group decreased the number of synovial in-flammatory cell infiltration and pus hyperplasia on day 30, and the number of arthritic claws[(1.50 ± 0.55)]was lower than that of Mgroup[(3.83±0.41)](P<0.05),the severity score of right posterior paw arthritis [(1.00 ± 1.10) points] was lower than that of M group[(2.83± 0.75) points](P<0.05), the mean score of limb arthritis [(0.75±0.27) points] was lower than [(2.17 ± 0.38) points](P<0.05),the relative ex-pression of CD68 (0.632 ± 0.017) was lower than that of M group (0.964±0.080) (P<0.05), the relative expression of iNOS (0.572± 0.036) was lower than that of M group (3.83 ± 0.41)(P<0.05).Conclusion Xinfeng capsule ameliorates arthrities-induced synovial in-jury in CIA mice, possibly by inhibiting m1-type polarization of macrophages, thereby reducing the release of inflammatory factors.
