目的探讨坤泰胶囊对围绝经期大鼠血管内皮损伤及卵巢组织中血管内皮生长因子( VEGF)和胰岛素样生长因子 -1(IGF-1)表达的影响。方法于 2017年 5月至 2018年 1月将 40只大鼠按随机数字表法分为对照组( n=10)、坤泰胶囊低剂量组(L-KT组, n=10)、坤泰胶囊中剂量组( M-KT组, n=10)、坤泰胶囊高剂量组( H-KT组, n=10)。对照组给予大鼠灌胃对照溶液; L-KT组、 M-KT组、 H-KT组大鼠分别给予坤泰胶囊灌胃 1.0 g/kg、2.0 g/kg、4.0 g/kg,每天一次,连续 12 w。采用逆转录聚合酶链式反应( RT-PCR)、蛋白质印迹法检测卵巢组织中 VEGF、IGF-1的 mRNA及蛋白表达。体外培养大鼠卵巢颗粒细胞,分别用含有 5 μM、2 0μM、50 μM坤泰胶囊的 H2O2或 H2O2处理,用放射免疫法测定雌二醇、孕酮、卵泡刺激素( FSH)和黄体生成素( LH)的水平。结果对照组大鼠卵巢呈现明显的衰老特征,表现为卵泡储备耗竭,卵巢间质致密,卵巢基质比例明显增加。与对照组相比, L-KT组、 M-KT组、 H-KT组大鼠的各级卵泡数目都有一定程度的恢复,结构趋于改善。与对照组相比, L-KT组、 M-KT组、 H-KT组大鼠外周血雌二醇水平逐渐升高(P<0.05)FSH、LH水平显著下降(P<0.05)。 RT-PCR检测结构显示,对照组、 L-KT组、 M-KT组、 H-KT组 VEGF mRNA相对表达量分别为1,.0±0.1、1.6±0.2、2.3±0.4、3.1±0.4,IGF-1 mRNA相对表达量分别为 1.0±0.1、1.4±0.1、2.1±0.3、2.8±0.5,H-KT组 VEGF mRNA、IGF-1 mRNA相对表达量均显著高于其他组。蛋白质印迹法检测结果与 RT-PCR结果趋势一致。与对照组相比, (400 μM)组细胞存活率下降约 40%(P<0.01)。然而,当 H2O2与不同浓度的坤泰 |

Objective To investigate the effects of Kuntai capsule on vascular endothelial injury and the expression of vascular endothelial growth factor (VEGF) and insulin-like growth factor-1 (IGF-1) in ovarian tissues of perimenopausal rats.Methods From May 2017 to January 2018, 40 rats were divided into a control group (n=10), Kuntai capsule low-dose group (L-KT group, n=10), medium-dose group (M-KT group, n=10) and high-dose group (H-KT group, n=10). The control group was given control solution by gavage; the rats in the L-KT group, M-KT group and H-KT group were given Kuntai capsules by gavage at 1.0 g/kg, 2.0 g/kg and 4.0 g/kg once a day for 12 w. The mRNA and protein levels of VEGF and IGF-1 in ovarian tissues were detected by reverse transcription polymerasechain reaction (RT?PCR) and Western blotting. Rat ovarian granulosa cells were cultured in vitro and treated with H2O2 or H2O2 containing 5 μM, 20 μM and 50 μM Kuntai capsules, and the levels of estradiol, progesterone, follicle-stimulating hormone as shown by depletion of follicle reserve, dense ovarian stroma and a significant increase in the proportion of ovarian stroma. Comparedwith the control group, the number of follicles at all levels in the L-KT group, M-KT group and H-KT groups was restored to some extent, and the structure tended to improve. Compared with the control group, the peripheral blood estradiol levels of rats in the L-KT group, M-KT group and H-KT groups gradually increased (P<0.05), and the levels of FSH and LH significantly decreased (P < 0.05). The RT?PCR results showed that the relative expression of VEGF mRNA in the control, L-KT, M-KT, and H-KT groups was 1.0±0.1, 1.6±0.2, 2.3±0.4, and 3.1±0.4, respectively, and the relative expression of IGF-1 mRNA was 1.0±0.1, 1.4±0.1, 2.1±0.3, and 2.8±0.5, respectively. The relative expression of VEGF mRNA and IGF-1 mRNA in the H-KT group was significantly higher than that in the other groups. The Western blotting results were consistent with the trend of the RT?PCR results. The cell survival rate of the H2O2 (400 μM) group was decreased by approximately 40% compared with that of the control group (P<0.01). However, when H2O2 was combined with different concentrations of Kuntai capsules (5 μM, 20 μM and 50 μM), we found that 20 μM and 50 μM Kuntai capsules could rescue the H2O2 -induced cell injury (P<0.001), while the estrogen secretion of cells in the Kuntai capsule-treated group was reduced by approximately 39% after H2O2 treatment compared with the control group (P<0.001).Conclusion High-dose Kuntai capsule had themost significant protective effect on vascular function in perimenopausal rats and could promote ovarian function by promoting the expression of VEGF and IGF-1 in ovarian cells. |
