| 目的建立高效液相色谱 -二极管阵列检测器( HPLC-DAD)法同时测定更年宁中 7种有效成分的含量。方法 2021年 6月至 2022年 2月,采用 C18色谱柱( 4.6 mm×250 mm,5 μm)以 0.1%磷酸水溶液 -乙腈作为流动相进行梯度洗脱,流速为 1.0 mL/min,柱温为 35 ℃,检测波长为 232 nm、226 nm、276 nm。结果,芍药苷、特女贞苷、黄芩苷、汉黄芩苷、丹皮酚、黄芩素、汉黄芩素分别在 6.24~103.96、8.21~136.80、14.48~241.36、2.29~38.10、3.33~55.46、1.16~19.36、0.43~7.22 mg/L范围内线性关系良好,相关系数 r≥0.999 5,平均回收率分别为 100.00%、99.30%、98.48%、97.26%、97.94%、102.01%、99.33%,相应的相对标准偏差(RSD)为 2.09%~4.05%。结论该法准确、可靠、灵敏度高,为进一步完善更年宁的质量标准提供依据。 |

| Objective To establish a high performance liquid chromatography-diode array detector (HPLC-DAD) method for simultaneous determination of 7 effective components in Gengnianning.Methods From June 2021 to February 2022, the chromatographic separation of HPLC-DAD method was achieved on a C18 column (4.6 mm×250 mm, 5 μm) with 0.1 % phosphoric acid solution-acetonitrile as mobile phase at the flow rate of 1.0 mL/min for gradient elution; The column temperature was 35 ℃, and the detection wavelengthswere 232 nm, 226 nm and 276 nm.Results The linear relationships of paeoniflorin, specnuezhenide, baicalin, wogonoside, paeony phenol, baicalein, wogonin were good within the ranges of 6.24-103.96, 8.21-136.80, 14.48-241.36, 2.29-38.10, 3.33-55.46, 1.16-19.36, 0.43-7.22 mg/L, respectively. The correlation coefficient was not less than 0.999 5, and the average recoveries were 100.00%, 99.30%,98.48%, 97.26%, 97.94%, 102.01% and 99.33%, respectively, with the RSDs ranging from 2.09% to 4.05%.Conclusion The method is accurate, reliable and sensitive. It provides technical reference for further improving quality standard of Gengnianning. |
