      目的检测宫颈癌病人外周血中 Th17细胞的比例及相关细胞因子白细胞介素( IL)-1β、IL-6的表达水平,探讨宫颈癌外周血中单核细胞对 Th17细胞分化的影响及机制。方法 2019年 6月至 2020年 12月在潍坊市人民医院妇科收治的宫颈癌病人(宫颈癌组) 39例,对照组( 30例)为健康志愿者及因良性疾病在潍坊市人民医院妇科就诊的病人,流式细胞仪检测宫颈癌组、对照组外周血中 Th17细胞的比例;实时荧光定量逆转录聚合酶链反应( qRT-PCR)检测宫颈癌病人、对照组外周血中 IL1β、IL-6的表达水平;分离健康志愿者外周血中的初始 CD4+T细胞,分别与对照组、宫颈癌组外周血中分离出的单核细胞进行共培养,并分别加入 anti-IL-1β、anti-IL-6中和抗体,流式细胞仪检测 Th17细胞的比例。结果宫颈癌组外周血中, Th17细胞比例( 3.5±1.4)%、IL-1β 0.457(0.078,1.193)、 IL-6 0.094(0.043,0.272)的表达水平明显高于对照组( 2.3±1.6)%、0.279(0.017,0.686)、 0.038(0.019,0.112)。初始 CD4+T细胞与宫颈癌组外周血中的单核细胞共培养, Th17细胞分化的比例( 1.77±0.61)%显著高于对照组( 0.90±0.39)%;与对照组相比(1.82±0.47)%,加入中和抗体后, Th17细胞的分化比例均下降,且加入 anti-IL-1β组(0.76±0.23)%与加入 anti-IL-6(1.06±0.34)%组比,前者 Th17细胞分化比例下降更为明显。结论宫颈癌病人外周血中的单核细胞更能促进 Th17细胞的分化。在 Th17细胞分化的过程中, IL-1β、IL6起到重要作用,且 IL-1β在促进 Th17细胞分化的过程中诱导作用更强。

      Objective To explore of the effect of monocytes in peripheral blood of cervical cancer on the differentiation of Th17cells by assessing the ratio of Th17 cells in cervical cancer patients(CC), the expression levels of relevant cytokines IL-1β and IL-6, and by changing the concentrations of IL-1β and IL-6 secreted by monocytes in the microenvironment.Methods Flow cytometry wasused to detect the proportions of Th17 cells in the peripheral blood of CC patients and the healthy control group (HC). qRT-PCR was applied to detect the expression levels of IL-1β and IL-6 in CC patients and the HC group. The isolated initial CD4+T cells from the peripheral blood of the HC group were respectively co-cultured with monocytes isolated from the HC group and CC patients, and different neutralizing antibodies were added, such as anti-IL-1β, anti-IL-6 and anti-IL-1β+anti-IL-6. The ratio of Th17 cells was measured by flow cytometry.Results In the peripheral blood of CC patients, the ratio of Th17 cells (3.5±1.4) % and the concentrations of IL-1β0.457 (0.078, 1.193) and IL-6 0.094 (0.043, 0.272) were markedly increased compared with those of the HC group (2.3±1.6) %, 0.279(0.017, 0.686), 0.038 (0.019, 0.112). The initial CD4+ T cells were respectively cultured together with monocytes from the peripheralblood of CC patients and HC group, higher Th17 cells ratio was observed in CC patients (1.77±0.61) % vs. (0.90±0.39) %. After addingneutralizing antibodies, the differentiation ratio of Th17 cells decreased compared with the control group (1.82±0.47) %, and when added to the anti-IL-1β group (0.76±0.23) %, the differentiation ratio of Th17 cells decreased more significantly than the anti-IL-6 group (1.06±0.34) % . Conclusions The monocytes in the peripheral blood of CC patients can significantly promote the differentiation of Th17 cells. IL-1β and IL-6 both play an important role in the process of Th17 cell differentiation, and IL-1β has a stronger inducing effect than IL-6.
