      目的考察自主研发的协定方中药“金贯蓝”提取物对寻常痤疮三种主要致病菌(痤疮丙酸杆菌、表皮葡萄球菌、金黄色葡萄球菌)的体外抑菌效果,为其临床治疗寻常痤疮提供一定实验依据。方法 2021年 1月至 2022年 3月采用琼脂打孔法考察金贯蓝提取物对三种痤疮相关致病菌的抑菌活性,并评估不同浓度药物对上述菌的抗菌作用是否具有浓度相关性;采用倍比稀释法测定药物最低抑菌浓度以及最小杀菌浓度,绘制药物时间抑菌曲线进一步考察其作用特点。结果 500 g/L金贯蓝提取物对 C. acnes、S. epidermidis、S. aureus的抑菌圈直径分别为( 27.67±2.52)mm、(23.33±1.53)mm、(22.00±1.00)mm;浓度为 250 g/L金贯蓝提取物对 C. acnes、S. epidermidis、S. aureus的抑菌圈直径分别为( 22.33±0.58)mm、(20.67±1.53)mm、(20.11±1.20) mm;浓度为 100 g/L金贯蓝提取物对 C. acnes、S. epidermidis、S. aureus的抑菌圈直径分别为( 13.30±0.75)mm、(10.67±1.53)mm、(10.67±0.58)mm,抑菌圈直径与提取物浓度呈正相关关系( r>0.90,P<0.001)。金贯蓝提取物对痤疮丙酸杆菌、表皮葡萄球菌、金黄色葡萄球菌的最小抑菌浓度均为 25 g/L;最小杀菌浓度分别为 50 g/L、50 g/L、25 g/L。抑菌曲线表明其主要影响细菌生长的迟缓期。结论中药金贯蓝提取物对寻常痤疮三种主要致病菌(痤疮丙酸杆菌、表皮葡萄球菌和金黄色葡萄球菌)有较好的抑制作用,且与药物浓度呈正相关,值得进一步开发研究。
英文摘要:
      Objective To investigate the effect of Jinguanlan extract on the three main pathogenic bacteria of acne vulgaris (Cutibac? terium acnes, Staphylococcus epidermidis and Staphylococcus aureus), so as to provide a certain experimental basis for its clinical treatment of acne vulgaris.Methods From January 2021 to March 2022, Agar drilling method was used to investigate the antibacterial activity of the extract against three acne-related pathogenic bacteria, and to evaluate the concentration correlation of the antibacterial effect ofdifferent concentrations of drugs on the above-mentioned bacteria. The minimum inhibitory concentration and minimum bactericidalconcentration were determined by multiple dilution method, and the time inhibition curve was drawn to further investigate its action characteristics.Results The bacteriostatic zone diameters of 500 g/L Jinguanlan extract against C. acnes, S. epidermidis and S. aureus were (27.67±2.52) mm, (23.33 ±1.53) mm and (22.00±1.00) mm, respectively. The bacteriostatic zone diameters of 250 g/L Jinguanlan extract against C. acnes, S. epidermidis and S. aureus were (22.33 ±0.58) mm, (20.67 ±1.53) mm and (20.11 ±1.20) mm, respectively. Thebacteriostatic zone diameter of 100 g/L the extract against C. acnes, S. epidermidis and S. aureus were (13.30±0.75) mm, (10.67±1.53)mm and (10.67±0.58) mm, respectively. There was a positive correlation between the diameter of the antibacterial zone and the concentration of the extract (r>0.90 P<0.001). The MIC of Cutibacterium acnes, Staphylococcus epidermidis, and Staphylococcus aureus were all 25 g/L; MBC was 50 g/L, 50 g/L, 25 g/L, respectively. The bacteriostatic curve showed that it mainly affected the slow period of bacterialgrowth.Conclusion The extract of JINGUANLAN formula has a good inhibitory effect on the three main pathogenic bacteria of acnevulgaris, and it is positively correlated with the drug concentration, which is worthy of further development and research.
