      目的探究泛素特异性蛋白酶 18(USP18)、干扰素刺激基因 15(ISG15)在乳腺癌组织中的表达水平及临床意义。方法采用 Ualcan数据库分析 USP18、ISG15在正常乳腺组织和乳腺癌组织中的表达情况及二者与乳腺癌预后的关系;选取渭南市中心医院 2012年 5月至 2015年 7月诊治的 99例乳腺癌病人癌组织、癌旁正常组织进行研究。分别检测 USP18、ISG15 mRNA及其蛋白表达情况;分析乳腺癌组织 USP18、ISG15表达水平与病人临床病理特征、预后的关系; Cox回归分析乳腺癌病人预后的影响因素。结果 Ualcan数据库中乳腺癌组织 USP18为 18.40±4.17、ISG15 mRNA表达水平为 168.56±43.95高于正常乳腺组织( 10.00±3.14、30.76±8.23)(P<0.05);乳腺癌组织 USP18、ISG15 mRNA表达水平高低与乳腺癌预后无明显相关性。乳腺癌组织 USP18为 1.67±0.53、ISG15 mRNA为 1.86±0.61及蛋白阳性表达率均高于癌旁正常组织( 1.03±0.34、0.99±0.33)(P<0.05);乳腺癌组织 USP18、ISG15表达水平均与淋巴结转移、乳腺癌分子亚型、肿瘤分化程度、 TNM分期相关( P<0.05);乳腺癌病人癌组织中 USP18表达水平与 ISG15呈正相关( P<0.05); USP18阳性组、 ISG15阳性组乳腺癌病人术后 60个月生存率均低于 USP18阴性组、 ISG15阴性组(P<0.05);淋巴结转移、 TNM分期、 USP18、ISG15均是影响乳腺癌病人不良预后的独立危险因素(P <0.05)。结论乳腺癌病人癌组织 USP18、ISG15表达水平均较高,两者均与乳腺癌病人预后关系密切,检测乳腺癌组织 USP18、ISG15水平有助于评估乳腺癌病人预后。

      Objective To investigate the expression levels and clinical significance of ubiquitin-specific protease 18 (USP18) and interferon stimulated gene 15 (ISG15) in breast cancer tissues. Methods Ualcan database was used to analyze the expression ofUSP18 and ISG15 in normal breast tissue and breast cancer tissue and their relationship with breast cancer prognosis; the cancer tissueand normal tissue adjacent to cancer of 99 cases of breast cancer patients diagnosed and treated in Weinan Central Hospital duringMay 2012 to July 2015 were selected for study. The mRNA and protein expression of USP18 and ISG15 was detected, respectively; therelationship between the expression levels of USP18 and ISG15 in breast cancer tissues and clinicopathological features and prognosiswas analyzed. Cox regression analysis was used to analyze the prognostic factors of breast cancer patients.Results The expression levels of USP18 (18.40±4.17) and ISG15 mRNAs (168.56±43.95) in breast cancer tissues in Ualcan database were higher than those innormal breast tissues [(10.00±3.14), (30.76±8.23)] (P<0.05); there is no significant correlation between USP18 and ISG15 mRNA expression levels in breast cancer tissue and breast cancer prognosis. The mRNA expression levels and the positive expression rates ofUSP18 (1.67±0.53) and ISG15 (1.86±0.61) in breast cancer tissues were higher than those in normal tissues adjacent to cancer [(1.03±0.34), (0.99±0.33)] (P<0.05); the expression levels of USP18 and ISG15 in breast cancer tissues were correlated with lymph node metastasis, breast cancer molecular subtype, tumor differentiation, and TNM staging (P<0.05); there was a positive correlation between the expression level of USP18 and ISG15 in cancer tissues of breast cancer patients (P<0.05); the 60-month survival rates of breast cancer patients in USP18 positive group and ISG15 positive group were significantly lower than those in USP18 negative group and ISG15 negative group (P<0.05); lymph node metastasis, TNM staging, USP18, ISG15 were independent risk factors for poor prognosis of breast cancer patients (P<0.05).Conclusion The expression levels of USP18 and ISG15 in breast cancer tissues of breast cancer patients arehigher, which are closely related to the prognosis of breast cancer patients. Detection of USP18 and ISG15 levels in breast cancer tissues is helpful to evaluate the prognosis of breast cancer patients.
