      目的探讨腹腔注射 α-半乳糖苷神经酰胺( α-GalCer)对小鼠肺恒定自然杀伤 T细胞( iNKT)、细胞亚群及外周血细胞因子的影响,从而为治疗相关肺部疾病提供基础研究数据。方法 60只 6~8周龄雄性 C57BL/6小鼠,采用随机数字表法分为 α-GalCer干预组和正常对照组,每组 30只,选择不同时间点观察干预组与对照组小鼠肺 iNKT细胞、亚群及外周血细胞因子的变化。结果组间比较发现,与对照组( 3.49±0.25)%相比,腹腔注射 α-GalCer可引起肺脏 iNKT细胞频率及亚群的变化,差异有统计学意义( P<0.05)。 α-GalCer干预组肺 iNKT细胞频率前 2d(6.56±0.05)%明显升高而后[d6(2.56±0.08)%、d8(2.64±0.14)%、d10(2.37±0.24)%]降低( P<0.05)。 iNKT 1细胞频率低于对照组且呈降低的趋势(P<0.05)。 iNKT 2细胞频率高于对照组在第 4天肺部 iNKT2细胞频率达到峰值,随后降低( P<0.05)。iNKT 17细胞频率略高于对照组,差异无统计学意义( P>0.05),与第 2天相比,第 4、6、8、10天 iNKT17细胞频率均显著升高( P<0.05)。组间比较发现,干预组与对照组外周血血清 IFN-γ、IL4、IL-17A浓度比较,差异有统计学意义, α-GalCer干预组在不同时间点 IFN-γ、IL-4、IL-17A浓度比较,差异有统计学意义,与第 2天相比, IFN-γ、IL-4水平于第 6天、第 8天均降低( P<0.05);IL-17A水平于第 6天降低( P<0.05)。α-GalCer干预组的 IFN-γ水平与对照组相比均有明显升高,差异有统计学意义( P<0.05);除第 8天外, α-GalCer干预组的 IL-4水平与对照组比较,差异有统计学意义( P<0.05);第 2天、第 8天 α-GalCer干预组的 IL-17A水平均低于对照组,差异有统计学意义( P<0.05)。结论腹腔注射 α-GalCer对小鼠肺组织 iNKT细胞和亚群有不同的激活效应; iNKT细胞激活后,会由脉管系统迁入间质;腹腔注射 α-GalCer可诱导血清细胞因子的变化,提示 α-GalCer注射可影响机体免疫功能。

      Objective To explore the effects of α-galactosylceramide (α-GalCer) treatment at different time on iNKT cells, cell subsets and peripheral blood cytokines of mouse lung, so as to provide basic research data for the treatment of related lung diseases.Meth ods Sixty C57BL/6 male mice aged 6-8 weeks were randomly divided into α-GalCer intervention group and control group, with 30mice in each group. Changes of iNKT cells, cell subsets and cytokines in peripheral blood of mice in α-GalCer intervention group and control group were observed at different time.Results Comparison between groups found that intraperitoneal injection α-GalCer could cause changes of lung iNKT cells frequency and subsets compared with the control group (3.49±0.25) %, and the difference was statistically significant (P<0.05). The frequency of lung iNKT cells increased significantly in the first 2 days (6.56±0.05) % and then decreased [d6 (2.56±0.08) %, d8 (2.64±0.14) %, d10 (2.37±0.24) %] (P<0.05). The frequency of lung iNKT 1 cells in α-GalCer interven tion group was lower than that in the control group (P<0.05). The iNKT 2 cell frequency was higher than the control lung, iNKT2 cellfrequency peaked on day 4 and subsequently decreased (P<0.05). The iNKT 17 cell frequency was slightly higher than that of the con trol group, the difference was not statistically significant (P>0.05), and the iNKT17 cell frequency was significantly higher on days 4, 6, 8, and 10 compared to day 2 (P<0.05). Comparison between groups found that peripheral serum IFN-γ, IL-4 and IL-17A showed IFN-γ, IL-4 and IL-17A concentrations at different time points, IFN-γ and IL-4 levels decreased on day 6 and 8 (P<0.05) and I L-17A levels decreased on day 6 (P<0.05). IFN-γ levels in α-GalCer intervention compared with the control group (P<0.05), IL-4 levels except day 8 in α-GalCer ntervention (P<0.05), and IL-17A was lower than the control group (P<0.05).Conclusions Intraperitoneal injection of α -GalCer has different activation effects on iNKT1 cells and subsets in mouse lung tissue. iNKT cells migrate from the vasculature to theinterstitium after activation. The intraperitoneal injection of α-GalCer can induce changes in serum cytokines, suggesting that α-GalCer injection can affect the body's immune function.
