目的基于网络药理学技术探索二白膏治疗慢性湿疹的分子机制。方法 2022年 1―5月通过 TCMSP数据库及《中国药典( 2015版)》筛选二白膏主要活性成分并通过 SwissTargetPrediction网站进行靶点预测;同时从 OMIM数据库、 GeneCards数据库筛选慢性湿疹的相关靶点;成分靶点与疾病靶点映射后使用 String网站及 Cytoscape 3.9.1软件构建蛋白质 -蛋白质相互作用网络,并采用分子对接技术对潜在活性成分与关键靶点进行匹配;通过 DAVID网站进行 GO与 KEGG分析; Cytoscape3.9.1软件构建药材 -成分 -靶点 -通路网络图。结果从二白膏中筛选出 17个活性成分和 97个潜在靶点,其中 15个核心靶点 18条通路与其治疗慢性湿疹有密切关系。结论二白膏治疗慢性湿疹具有生物学支撑。不同的蛋白存在协同作用,其中 AKT1、 SRC和 EGFR等靶点起着关键作用。生物过程主要涉及细胞因子受体结合、炎症反应、免疫反应等,并主要富集在 PI3K-Akt、 JAK-STAT信号通路上,发挥抗炎、抗氧化、调控细胞增殖和凋亡、修复皮肤屏障等作用。 |

Objective To explore the molecular mechanism of Erbai ointment in the treatment of chronic eczema based on network pharmacology.Methods The main active components of Erbai ointment were screened by TCMSP database and Chinese Pharmacopoeia (2015 edition) and the target was predicted by SwissTargetPrediction website from January to May 2022. At the same time, the related targets of chronic eczema were screened from OMIM database and GeneCards database. After mapping between component targets and disease targets, protein-protein interaction network was constructed by String website and Cytoscape 3.9.1 software, and molecular docking technology was used to match potential active components with key targets. GO and KEGG were analyzed by DAVID website. Cytoscape 3.9.1 software was used to construct herb-component-target-pathway network.Results 17 active components and 97 potential targets were screened from Erbai ointment, of which 15 core targets and 18 pathways were closely related to the treatment of chronic eczema.Conclusion Erbai ointment has biological support in the treatment of chronic eczema. Different proteins have synergistic effects, among which AKT1, SRC and EGFR etc. play a key role.The biological process mainly involves cytokine receptor binding, inflammatory response, immune response etc. These targets are mainly concentrated in multiple signaling pathways such as PI3KAkt and JAK-STAT signal pathway, which play key roles of anti-inflammatory, antioxidant, regulation of cell proliferation and apopto |
