目的探究子宫内膜癌病人血清长基因间非编码 RNA00511(LINC00511)微 RNA-515-5p(miR-515-5p)表达水平与临床病理特征及预后的关系。方法收集 2017年 3月至 2019年 2月海南医学院第二附、属医院子宫内膜癌病人(子宫内膜癌组) 82例;同时收集子宫内膜不典型增生病人(不典型增生组) 67例及健康体检者(对照组) 79例。采用实时荧光定量逆转录聚合酶链式反应( qRT-PCR)检测血清 LINC00511、miR-515-5p表达水平;采用 Pearson相关分析子宫内膜癌病人血清 LINC00511与 miR-515-5p表达水平的相关性;采用 Kaplan-Meier法分析子宫内膜癌病人血清 LINC00511、miR-515-5p表达水平与预后的关系;多因素 Cox回归分析影响子宫内膜癌病人预后的因素。结果对照组、不典型增生组、子宫内膜癌组血清 LINC00511表达水平( 1.02±0.21、1.82±0.31、2.60±0.45)依次升高, miR-515-5p表达水平( 1.01±0.20、0.70±0.18、0.42±0.16)依次降低( P<0.05)。肌层浸润深度 ≥1/2、有淋巴结转移、临床分期 Ⅲ~Ⅳ期的子宫内膜癌病人血清 LINC00511表达水平高于肌层浸润深度 <1/2、无淋巴结转移、临床分期 I~Ⅱ期病人, miR-515-5p表达水平低于肌层浸润深度 <1/2、无淋巴结转移、临床分期 Ⅰ~Ⅱ期病人( P<0.05)。子宫内膜癌病人血清 LINC00511与 miR-515-5p表达水平呈负相关( r= 0.48,P<0.05)。子宫内膜癌病人血清 LINC00511高表达病人 3年生存率低于 LINC00511低表达病人( P<0.05); miR-515-5p高表达病人 3年生存率高于 miR-515-5p低表达病人( P<0.05)。 LINC00511、临床分期是子宫内膜癌病人 3年内死亡的独立危险因素( P<0.05),miR-515-5p是子宫内膜癌病人 3年内死亡的保护因素(P<0.05)。结论子宫内膜癌病人血清 LINC00511表达上调, miR-515-5p表达下调,二者均与肌层浸润深度、淋巴结转移、临床分期及预后有关。 |

Objective To investigate the relationship between the expression levels of serum long intergenic non-coding RNA 00511 (LINC00511) and microRNA-515-5p (miR-515-5p) with clinicopathological features and prognosis of patients with endometrial cancer.Methods Eighty-two patients with endometrial cancer (endometrial cancer group) in the Second Affiliated Hospital of HainanMedical University from March 2017 to February 2019 were collected. Meanwhile, 67 patients with atypical endometrial hyperplasia(atypical hyperplasia group) and 79 healthy people (control group) were collected. QRT-PCR method was performed to detect the ex pression levels of serum LINC00511 and miR-515-5p; Pearson correlation was performed to analyze the correlation between serum LINC00511 and miR-515-5p expression levels in patients with endometrial cancer; Kaplan-Meier method was performed to analyze the relationship between the expression levels of serum LINC00511 and miR-515-5p and prognosis in patients with endometrial cancer;multivariate Cox regression was performed to analyze the prognostic factors in patients with endometrial cancer.Results The expression level of serum LINC00511 (1.02±0.21, 1.82±0.31, 2.60±0.45) in the control group, the dysplasia group and the endometrial cancergroup was increased sequentially, and the expression level of miR-515-5p (1.01±0.20, 0.70±0.18, 0.42±0.16) was decreased sequential ly (P<0.05). The expression level of serum LINC00511 in endometrial cancer patients with myometrial invasion depth ≥ 1/2, lymphnode metastasis and clinical stage Ⅲ-Ⅳ was higher than that in patients with myometrial invasion depth<1/2, no lymph node metasta sis and clinical stage Ⅰ-Ⅱ, and the expression level of miR-515-5p was lower than that in patients with myometrial invasion depth<1/ 2, no lymph node metastasis and clinical stage Ⅰ-Ⅱ (P<0.05). Serum LINC00511 in patients with endometrial cancer was negatively correlated with the expression level of miR-515-5p (r=0.48, P<0.05). The 3-year survival rate of patients with high serum LINC00511expression in patients with endometrial cancer was lower than that in patients with low LINC00511 expression (P<0.05); the 3-year sur vival rate of patients with high miR-515-5p expression was higher than that of patients with low miR-515-5p expression (P<0.05). LINC00511 and clinical stage were independent risk factors for death within 3 years in patients with endometrial cancer (P<0.05), and miR-515-5p was a protective factor for death within 3 years in patients with endometrial cancer (P<0.05). Conclusion Serum LINC00511 expression is up-regulated and miR-515-5p expression is down-regulated in patients with endometrial cancer, both of themare related to the depth of myometrial invasion, lymph node metastasis, clinical stage and prognosis. |
