      目的基于网络药理学的方法研究头花蓼对幽门螺杆菌胃炎以及胃癌的药理学机制,其中槲皮素、没食子酸和槲皮苷是头花蓼的主要活性成分。方法通过 PubChem、STITCH、SEA等 8个数据库获取相关数据,并使用 Cytoscape软件绘制蛋白 蛋白相互作用及头花蓼 -化合物 -预测靶点网络图。利用 DAVID数据库对相关靶点进行 GO和 KEGG的富集分析,并应用 AutoDock软件进行分子对接,以分析 hub基因与其对应小分子的结合能。结果网络药理学分析结果表明头花蓼对幽门螺杆菌胃炎以及胃癌的作用靶点共有 27个,其中 degree值前 6的核心靶点和其相对应的化合物分子对接的结合能均小于 0,且 80%的最低结合能结果 ≤?5.0 kJ/mol。GO和 KEGG的富集结果表明头花蓼可以调控炎症反应、细胞凋亡信号通路、 TNF信号通路等生物学过程,能通过影响受体复合物、蛋白质异二聚体活性发挥作用。结论头花蓼多靶点、多途径地作用于幽门螺杆菌胃炎以及胃癌,为头花蓼老药新用提供了新的依据。
英文摘要:
      Objective The pharmacological mechanism of Polygonum capitatum on Helicobacter pylori gastritis and gastric cancerwas studied based on network pharmacology, in which quercetin, gallic acid and quercitrin were the main active components of Polygo? num capitatum.Methods Relevant data were obtained from 8 databases, including PubChem, STITCH, and SEA, and Cytoscape software was used to draw protein?protein interactions and network diagrams of the Polygonum capitatum-compound-predicted targets. TheDAVID database was used to conduct enrichment analysis of GO and KEGG for related targets, and AutoDock software was used formolecular docking to analyze the binding energy of hub genes and their small corresponding molecules.Results The results of the network pharmacology analysis showed that there were 27 targets of Polygonum capitatum on Helicobacter pylori gastritis and gastric cancer, of which the top 6 core targets of degree value and their corresponding compound molecular docking binding energies were all lessthan 0 and 80% minimum binding energy results ≤?5.0 kJ/mol. The GO and KEGG enrichment results suggested that Polygonum capi? tatum could regulate biological processes such as the inflammatory response, apoptosis signaling pathway and TNF signaling pathwayand could play a role by affecting the activity of receptor complexes and protein heterodimers.Conclusion Polygonum capitatum acts on Helicobacter pylori gastritis and gastric cancer with multiple targets and pathways, which provides a new basis for the new use of the old medicine Polygonum capitatum.
