目的探究血清及肝癌组织中长链非编码 RNA(LncRNA)人去泛素化酶含有卵巢肿瘤结构域的 6B反义 RNA1(OTUD6B-AS1)、微 RNA-365a-3p(miR-365a-3p)的表达水平与肝癌病人临床病理特征及预后的关系。方法纳入 2015年 3月至 2017年 3月海南医学院第一附属医院收治的肝癌病人 86例(肝癌病人组)同时募集同期体检的健康志愿者 86例(健康对照组),收集所有研究对象的临床资料。采用 qRT-PCR法检测血清及肝癌组织中,LncRNA OTUD6B-AS1、miR-365a-3p表达; Starbase数据库预测 LncRNA OTUD6B-AS1与 miR-365a-3p之间的关系;使用 Pearson分析血清中 LncRNA OTUD6B-AS1与 miR365a-3p表达的相关性;绘制 Kaplan-Meier生存曲线分析血清中 LncRNA OTUD6B-AS1、miR-365a-3p表达与病人 5年生存率之间的关系; Cox回归分析肝癌病人预后的影响因素。结果与健康对照组比较,肝癌病人组血清中 LncRNA OTUD6B-AS1高表达( 1.59±0.41比 1.02±0.32)miR-365a-3p低表达( 0.42±0.15比 1.05±0.26)(P<0.05)。肝癌组织中 LncRNA OTUD6B-AS1表达水平高于癌旁组织( 1.70±0.44比,0.97±0.16),miR-365a-3p表达水平低于癌旁组织( 0.53±0.17比 1.01±0.14)(P<0.05)。血清中 LncRNA OTUD6B-AS1、miR-365a-3p表达与 TNM期、淋巴结转移以及分化程度相关( P<0.05)。生物信息学预测显示 LncRNA OTUD6B-AS1与 miR-365a-3p之间存在靶向结合位点。 Pearson分析显示,肝癌病人血清中 LncRNA OTUD6B-AS1与 miR-365a3p表达存在负相关( r=?0.47,P<0.05)。 LncRNA OTUD6B-AS1低表达组肝癌病人的 5年生存率高于 LncRNA OTUD6B-AS1高表达组( 37.21%比 13.95%,χ2=6.11,P=0.013); miR-365a-3p高表达组肝癌病人的 5年生存率高于 miR-365a-3p低表达组(41.86%比 9.30%,χ2=8.03,P=0.005)。 LncRNA OTUD6B-AS1高表达以及 miR-365a-3p低表达是影响肝癌病人死亡的独立危险因素( P<0.05)。结论 LncRNA OTUD6B-AS1、miR-365a-3p在肝癌病人血清及肝癌组织中分别呈高、低表达,二者均与 TNM期、淋巴结转移、分化程度等临床病理特征以及预后有关。 |

Objective To explore the relationship between the expression levels of LncRNA OTUD6B-AS1 and miR-365a-3p in serum and liver cancer tissue with the clinicopathological characteristics and prognosis of patients with liver cancer.Methods Eighty-six patients with liver cancer (liver cancer patient group) admitted to the First Affiliated Hospital of Hainan Medical College fromMarch 2015 to March 2017 were included, and 86 healthy volunteers (healthy control group) who underwent physical examination wererecruited, and the clinical data of all subjects were collected. The expression of LncRNA OTUD6B-AS1 and miR-365a-3p in serum and liver cancer tissue was detected by qRT-PCR; Starbase database was performed to predict the relationship between LncRNA OTUD6BAS1 and miR-365a-3p; the correlation between LncRNA OTUD6B-AS1 and miR-365a-3p expression in serum was analyzed by Pear‐son; the relationship between the expression of LncRNA OTUD6B-AS1 and miR-365a-3p in serum and the 5-year survival rate of patients was analyzed by drawing Kaplan-Meier survival curve; Cox regression was performed to analyze the factors influencing the prognosis of patients with liver cancer.Results Compared with the healthy control group, the serum LncRNA OTUD6B-AS1 was highly expressed in the liver cancer patient group (1.59±0.41 vs. 1.02±0.32), and the miR-365a-3p was lowly expressed (0.42±0.15 vs. 1.05± 0.26) (P<0.05). The expression level of LncRNA OTUD6B-AS1 in liver cancer tissues was higher than that in adjacent tissues (1.70±0.44 vs. 0.97±0.16), and the expression level of miR-365a-3p in liver cancer tissues was lower than that in adjacent tissues (0.53±0.17vs. 1.01±0.14) (P<0.05). The expressions of LncRNA OTUD6B-AS1 and miR-365a-3p in serum were correlated with TNM stage, lymph node metastasis and degree of differentiation (P<0.05). Bioinformatics prediction revealed a targeted binding site between LncRNA OTUD6B-AS1 and miR-365a-3p. Pearson analysis showed that there was a negative correlation between the expression of LncRNA OTUD6B-AS1 and miR-365a-3p in serum of patients with liver cancer (r=?0.47, P<0.05). The 5-year survival rate of liver cancer patients in the LncRNA OTUD6B-AS1 low expression group was higher than that in the LncRNA OTUD6B-AS1 high expression group (37.21% vs. 13.95%, χ2=6.11, P=0.013); the 5-year survival rate of liver cancer patients in the miR-365a-3p high expression group was higher than that in the miR-365a-3p low expression group (41.86% vs. 9.30%, χ2=8.03, P=0.005). High expression of LncRNA OTUD6B-AS1 and low expression of miR-365a-3p were independent risk factors for death in patients with liver cancer (P<0.05).Con? clusion LncRNA OTUD6B-AS1 and miR-365a-3p expresse high and low levels in serum and liver cancer tissue of patients with livercancer, respectively, and both are associated with clinicopathological features such as TNM stage, lymph node metastasis, degree of differentiation and prognosis. |
